摘要
靶序列富集多重PCR(target enriched multiplex PCR,Tem–PCR)作为一种新型的分子生物学高通量检测技术,通过靶序列富集、靶序列加标签以及超级引物扩增3个阶段实现对靶标序列的大量扩增。靶序列富集多重PCR具有独特的反应过程,克服了常规多重PCR扩增条件不兼容、扩增具有偏爱性等缺点,实现了对同一反应体系多种病原的同步检测,为临床病原鉴别诊断、基因分型、耐药基因等领域的研究提供了新思路,具有较大的发展前景。笔者对Tem–PCR的研究背景、反应原理、技术应用及试剂盒开发等进行综述,旨在为Tem–PCR的研究和发展提供参考依据,为中国应对公共卫生危机提供新思路。
Target enriched multiplex PCR(Tem–PCR), a novel technology applied in molecular biology, includes three reaction stages which are enriching, tagging and amplifying by super primers. The promising Tem–PCR method solves the incompatibility and preference during amplification in routine PCR and makes pathogeny synchronous detection in one reacting system come true, hence, it gives to a new clue for researches in clinical pathogenic differential diagnosis, genotyping and resistance genes. This paper summarized the situation of commercial kits, research background and reaction principle based on the review of the applications of Tem–PCR in domestic and abroad with the purpose to provide references for further research in the field and new ideas for dealing with public health crisis.
