飞行时间质谱法对羊绒羊毛纤维蛋白的鉴别原理 被引量:2

摘要 针对目前羊绒羊毛的检测需求,文章主要探究质谱技术对羊绒羊毛纤维蛋白的鉴别原理。基于蛋白质组学原理,通过还原法提取毛绒角蛋白并用胰蛋白酶对其进行处理,得到分子量较小的肽溶液,最后用MALDI-TOF MS进行检测。结果表明,羊绒区别于羊毛的肽段荷质比为2 691.4,羊毛区别于羊绒的肽段质荷比为2 664.5,通过质谱网络数据库检索得出这2种肽段的氨基酸排列顺序存在一个位点的差异,即在羊毛中是S(丝氨酸)的位置,在羊绒中变成了N(天冬酰胺),据此可以进行羊绒和羊毛的鉴别。 Aiming at the detection requirements of cashmere wool, we explore the mass spectrometry identification principle of cashmere wool fiber protein. On the basis of the principle of proteomics, we extract the wool keratin by reduction method and treatment of trypsin to get smaller molecular weight peptide solution, finally we detect the smaller molecular weight peptide solution by MALDI - TOF MS. Results show that cashmere peptide has 2 691.4 mass-to-charge ratio, which is different from wool, and the wool peptide mass-to-charge ratio is 2 664.5. By mass spectrometry of network database retrieval it is concluded that the two have different amino acid sequence of peptides, that is, S (serine) position in wool and N (asparagine) in cashmere, which can be used in the identification of cashmere and wool.
