摘要
用超声波辅助纤维素酶法提取芦荟多糖,并测定其抗肿瘤活性。采用响应面法优化超声波辅助纤维素酶提取芦荟多糖的条件。通过测定芦荟多糖对人体肝癌细胞HepG2生长的影响,研究其抗肿瘤活性。结果显示,当料液比1/30(g/mL)、超声波功率600W、pH5.0时,最佳提取条件为加酶量0.2%、提取温度49℃、提取时间16min。此条件下芦荟多糖的提取率为5.99%,比超声波辅助法和纤维素酶法分别提高了9.91%和37.38%。芦荟多糖具有较好的抗肿瘤活性,随着浓度的增加,其对HepG2细胞的生长抑制率逐渐增强。当其质量浓度为160mg/mL时,使70%的HepG2细胞处于裂解状态。超声波辅助纤维素酶法是一种新的、有效的芦荟多糖提取方法。
This study focused on the extraction of aloe polysaccharides by cellulase assisted with ultrasonic, and the antitumor activity of aloe polysacch-arides was also evaluated. The extraction conditions of aloe polysaccharides were optimized using response surface methodology, and the antitumor activity was assessed by measuring the effect of aloe polysaccharides on growth of human hepatoma cells Hep G2. Results showed that when solid-liquid ratio of 1/30(g/m L),ultrasonic power of 600 W and p H 4.5, the optimum extraction conditions were enzyme dosage of 0.2%, reaction temperature 49 ℃, and reaction time 16 min. Under such condition, polysaccharides extraction rate reached 5.99%, improved by 9.91% and 37.38% respectively, compared to treatment with ultrasonic or cellulose singly. Polysaccharides extracted showed good antitumor activity, with the increase of polysaccharides concentration, it exhibited higher inhibition rate on Hep G2 cells. Under the concentration of 160 mg/m L, about 70% of the Hep G2 cells was in lysis state. The method of cellulase assisted with ultrasonic is a new and effective way for aloe polysaccharides extraction, and aloe polysaccharides possess good antitumor activity.
