摘要
为了促进辣木组培快繁,以辣木种子为外植体建立无菌体系,并对其进行增殖及生根诱导研究。结果表明:辣木种子较好的灭菌条件为不剥皮+70%酒精(30s)+0.1%升汞(8min),其成活率达73.3%,污染率和灭死率分别为26.7%和0;最适的增殖培养基为MS+NAA0.01mg·L-1+6-BA0.4mg·L-1,增殖系数达3.9,且苗生长健壮;较好的生根培养基为1/2MS+IBA0.4mg·L-1,其生根率达92%,平均根数为6.5根,根粗达0.044mm。
In order to promote tissue culture fast propagation of Moringa oleifera,the seeds of Moringa oleifera Lam.were used as explants to establish their aseptic system,and the proliferation culture and rooting culture were carried out.The results showed that the optimal sterilization condition for the seeds of Moringa oleifera Lam.with seed capsule was 70%alcohol(30s)+0.1% mercuric chloride(8min),in which the survival rate reached 73.3%,the rate of pollution and death were repecetively 26.7% and 0;The optimal proliferation culture medium was MS+NAA0.01 mg·L-1+6-BA0.4 mg·L-1 among all the tested culture media,in which the proliferation index was 3.9,and the seedlings grew healthy and strong;And the optimal rooting culture medium was 1/2MS+IBA0.4 mg·L-1,in which the rooting rate was 92%,the average number of roots reached 6.5and root thickness was 0.044 mm respectively.
