麻黄连翘赤小豆汤对肝细胞性黄疸小鼠保肝退黄作用的研究 被引量:7

摘要 目的:观察麻黄连翘赤小豆汤(MLC)对四氯化碳(CC1_4)所致小鼠急性化学肝损伤引起的肝细胞性黄疸的保肝退黄作用,并初步探讨其作用机制。方法:将60只SPF级KM雄性小鼠,随机分为6组,正常组,模型组和联苯双酯组(150mg/kg),MLC低、中、高剂量组,每组10只。连续灌胃给药7d,末次给药1h后,除正常组外,其余5组采用0.1%CC1_4一次腹腔注射制造CC1_4急性肝损伤,建立肝细胞性黄疸模型。禁食不禁水16h后摘眼球取血测定小鼠血清中总胆红素(TBIL)、谷丙转氨酶(ALT)和谷草转氨酶(ALT)水平,取肝脏测定各组小鼠的肝脏指数,HE染色观察肝脏病理学变化。结果:与正常组比较,模型组肝脏指数、血清TBIL、ALT和ALT水平均显著升高(P<0.01);与模型组相比,MLC高剂量组的肝脏指数明显降低(P<0.05),TBIL、ALT和AST水平均显著降低(P<0.01),MLC低、中剂量组的ALT水平显著降低(P<0.01)。结论:MLC对CCl4急性肝损伤所致的肝细胞性黄疸小鼠具有明显保肝退黄作用,其机制可能与抑制脂质过氧化反应有关。 Objective: To observe the effects of liver protecting and jaundice removing of Mahuang Lianqiao Chixiaodou Decoction(MLC) in hepatocellular jaundice mice caused by acute chemical liver injury induced by carbon tetrachloride(CCl4) and to discuss its mechanism. Methods: Sixty SPF grade KM male mice were randomly divided into six groups, including a normal group, model group, bifendate group(150mg/kg), and low, medium and high dose of MLC group(0.14, 0.28, 0.56g/m L, respectively), 10 mice in each group. And all the mice were intragastrically administrated the corresponding agents for 7d, respectively. 1h after the last administration, 0.1% CCl4 was given to mice in all groups except the normal group in a single intraperitoncal injection to make the acute liver injury and to establish hepatocellular jaundice model. 16 h of fasting but water later, the mice were sacrificed for the blood from eyeballs, and the levels of total bilirubin(TBIL), alaninc aminotransfcrasc(ALT) and aspartatc aminotransfcrasc(AST) were detect, liver index was detected from livers, and HE staining was performed for observing the pathological changes of the liver tissue. Results: Compared with the normal group, the liver index, and the levels of serum TBIL, ALT, and AST in the model group were significantly increased(P<0.01); Compared with the model group, the liver index in high dose of MLC group was decreased significantly(P<0.05), and the levels of TBIL, ALT and AST were increased significantly(P<0.01), while the levels of ALT in the low and medium dose of MLC groups were increased significantly(P<0.01). Conclusion: MLC has a significant effect on liver protecting and jaundice removing in hepatocellular jaundice mice caused by acute chemical liver injury induced by CCl4, which might be related to the anti-oxidation.
