摘要
该试验以‘美人’梅离体培养的无菌苗叶片为材料 ,研究了不同种类激素配比对叶片脱分化及再分化的影响 ,并对不同条件下产生的愈伤组织进行了细胞学观察。结果表明 :BA对‘美人’梅叶片的愈伤组织诱导起到抑制作用 ;NAA可使叶片直接生根 ;2 ,4 D的加入可以诱发叶片形成球形愈伤。但只有配合适当浓度的BA ,才能形成胚状体。诱导胚状体合适的培养基为 :WPM +BA 1mg L +NAA 0 1mg L +2 ,4 D 1mg L +L 谷氨酰胺 10 0mg L +水解乳蛋白 10 0 0mg L。
Leaves of Mei (Prunus mume `Meiren' ) were employed as materials in this experiment. Influences of different hormones on the dedifferentiation and redifferentiation of leaves were investigated and cytological observations were carried out. The results show that BA restrains the induction of callus, NAA can directly induce rhizogenesis from leaves and 2,4--D can induce globular callus. Somatic embryogenesis appears only if 2,4--D and BA were used in the medium at the same time. The most suitable medium for somatic embryogenesis is WPM+BA 1 mg/L+NAA 0.1 mg/L+2,4--D 1 mg/L+L-glutamine 100 mg/L+casein hydrolyze 1 000 mg/L.
