摘要
目的探讨高良姜素对口腔癌CAL27细胞的生长和转移的影响。方法采用酶抑制动力学方法观察高良姜素对基质金属蛋白酶9(MMP-9)的抑制作用,分析不同浓度高良姜素对口腔癌CAL27细胞的毒理性、转移的抑制作用和CAL27细胞中TIMP-1、MMP-9、转化生长因子(TGF-β1)表达水平的影响。结果高良姜素能够抑制MMP-9的活性,诱导CAL27细胞的凋亡,但对正常细胞的毒性较低。随着高良姜素浓度的增加,口腔癌CAL27细胞中MMP-9和TGF-β1表达均呈降低趋势,TIMP-1表达呈升高趋势,且口腔癌CAL27细胞穿过滤膜的能力下降。结论高良姜素可通过下调MMP-9和TGF-β1的水平、增强TIMP-1表达,抑制口腔癌CAL27细胞的生长和转移。
Objective To investigate the effect of Galangin on CAL27 cell growth and metastasis of patients with oral carcinoma. Methods The inhibitory effect of Galangin on matrix metalloproteinase-9 (MMP-9) was studied using enzyme inhibition kinetics method, and the effects of Galangin with different concentrations on the inhibition of CAL27 cell growth and metastasis and on expressions of tissue inhibitor of metalloproteinase (TIMP-1) , MMP-9 and transforming growth factor-131 (TGF-β1) in CAL27 cells were also analyzed. Results Galangin could inhibit MMP-9 activity, induce CAL27 apoptosis with little toxicity for the normal cells. With the increasing concentration of Galangin, MMP-9 and TGF- 131 expression levels in CAL27 oral carcinoma cells were decreasing, and TIMP-1 expression level was increasing, and the ability of CAL27 oral carcinoma cells through artificial basement membrane was degraded. Conclusion Galangine may enhance TIMP-1 expression by inhibiting the MMP-9 and TGF-β1 levels, eventually playing an inhibitory role on CAL27 cell growth and metastasis.
