针康法对脑缺血大鼠脑组织VEGF、LN表达的影响 被引量:9

Expression of VEGF and LN in Brain Tissue of Ischemic Stroke Rats by Acupuncture- rehabilitation

摘要 目的:观察针康法对脑缺血大鼠脑组织血管内皮生长因子(VEGF)、层粘连蛋白(LN)表达的影响,探讨针康法促进神经功能恢复的相关作用机制。方法:建立大鼠永久性局灶性脑缺血模型,设立模型组、假手术组、针刺组、康复组及针康组。模型组、假手术组不予任何治疗,针刺组采用头穴丛刺针法治疗,康复组采用跑台训练模拟临床康复治疗,针康组采用针康法治疗。免疫组织化学法观察各时间点缺血脑组织VEGF、LN的动态变化。结果:术后7天、14天,与模型组相比,康复组、针刺组与针康组VEGF阳性表达增多(P<0.05);与康复组、针刺组比较,针康组VEGF阳性细胞数明显增多(P<0.05)。术后7天、14天,与模型组相比,康复组、针刺组与针康组LN阳性表达增多(P<0.05);与康复组、针刺组比较,针康组LN阳性细胞数明显增多(P<0.05)。结论:针康法可能通过上调缺血脑组织VEGF、LN的表达促进脑缺血后神经功能恢复。 Objective: To observe the expression of VEGF and LN in brain tissue of cerebral ischemia rats by acupuncture- rehabilitation,and to explore the mechanism of acupuncture- rehabilitation to promote the recovery of nerve function. Methods: The rat models of permanent focal cerebral ischemia were established. They were divided into model group,sham operation group,acupuncture group,rehabilitation group and acupuncture- rehabilitation group. Model group and sham operation group were without any treatment; the acupuncture group was treated by scalp acupuncture treatment; the rehabilitation group received treadmill training simulating clinical rehabilitation; acupuncture group was with acupuncture combined with rehabilitation. The changes of VEGF and LN in ischemic brain tissue at different time points were observed by immunohistochemistry. Results: After7 d,14 d compared with the model group and rehabilitation group,in the acupuncture group and acupuncture-rehabilitation group the expression of VEGF was increased( P〈0. 05); compared with the rehabilitation group and the acupuncture group,in the acupuncture- rehabiliation group the number of VEGF positive cells was increased significantly( P〈0. 05). After 7 d,14 d compared with the model group,rehabilitation group,acupuncture group and acupuncture- rehabilitation group the expression of LN was increased( P〈0. 05); compared with the rehabilitation group and acupuncture group,in the acupuncture- rehabilitation group the number of LN positive cells was increased significantly( P〈0. 05). Conclusion: Acupuncture- rehabilitation promotes the recovery of neural function after cerebral ischemia by increasing the expression of VEGF and LN in ischemic brain tissue.
