摘要
目的探讨缺氧诱导因子2α(HIF-2α)蛋白和血管内皮生长因子(VEGF)蛋白在宫颈鳞状细胞癌组织中的表达及意义。方法收集临床宫颈组织蜡块共208例,其中宫颈鳞状细胞癌102例,宫颈上皮内瘤样病变(CIN)66例,正常宫颈组织40例。通过免疫组织化学方法检测各组织中HIF-2α蛋白和VEGF蛋白的表达,并分析HIF-2α蛋白与宫颈鳞状细胞癌临床病理参数的关系,HIF-2α蛋白和VEGF蛋白表达的相关性及HIF-2α蛋白和VEGF蛋白的表达与微血管密度(MVD)的关系。结果 HIF-2α蛋白在正常宫颈组织、CIN和宫颈鳞状细胞癌中表达阳性率分别为0.0%、18.2%、73.5%,VEGF蛋白在正常宫颈组织、CIN和宫颈鳞状细胞癌中表达阳性率分别为7.5%、21.2%、68.6%;HIF-2α蛋白和VEGF蛋白在3种组织中的表达阳性率差异均有统计学意义(P<0.05)。HIF-2α蛋白在宫颈鳞状细胞癌组织中的表达与淋巴结转移、浸润深度和临床分期有关(P<0.05),而与年龄和病理组织学分化程度无关(P>0.05);HIF-2α蛋白和VEGF蛋白表达呈正相关(P<0.05);MVD在HIF-2α蛋白、VEGF蛋白阳性组的表达均高于其阴性组的表达(P<0.05)。结论 HIF-2α可能通过调控VEGF的表达而促进宫颈鳞状细胞癌的发生和发展,并可能成为宫颈鳞状细胞癌治疗的一个新的分子靶点。
Objective To explore the expressions of hypoxia-inducible factor 2α( HIF-2α) protein and vascular endothelial growth factor( VEGF) protein in cervical squamous cell carcinoma and their clinical significance. Methods Two hundred and eight paraffin blocks of cervical tissue were collected,including 102 cervical squamous cell carcinoma cases,66 cervical intraepithelial neoplasia( CIN) cases and 40 normal cervical tissue. The expressions of HIF-2α protein and VEGF protein in each tissue were detected by immunohistochemistry. The relationships between the positive expression of HIF-2α protein and clinicopathological parameters of cervical squamous cell carcinoma were analyzed; the correlation between the expression of HIF-2α protein and VEGF protein were analyzed; the relationships between the expression of HIF-2α protein,VEGF protein and micro vessel density( MVD) were analyzed,respectively. Results The positive expression rate of HIF-2α protein in normal cervical tissue,CIN and cervical squamous cell carcinoma tissue was 0. 0%,18. 2% and 73. 5% respectively. The positive expression rate of VEGF protein in normal cervical tissue,CIN and cervical squamous cell carcinoma tissue was 7. 5%,21. 2%and 68. 6% respectively. There was statistic difference of positive expression rate of HIF-2α protein and VEGF protein among the three tissues( P 〈 0. 05). The expression of HIF-2α protein was related to lymph node metastasis,deeper of invasion and clinical stage( P 〈 0. 05),while the expression of HIF-2α protein was not related to age and histological grade( P 〉 0. 05).There was positive correlation between the expressions of HIF-2α protein and VEGF protein in cervical squamous cell carcinoma( P 〈 0. 05). The expression of MVD in positive HIF-2α,VEGF group was significantly higher than that in negative HIF-2αand VEGF group( P 〈 0. 05). Conclusion HIF-2α may promote the occurrence and progression of cervical squamous cell carcinoma by regulating the expression of VEGF,which may provide a new therapeutic target for the treatment of cervical squamous cell carcinoma.
