摘要
目的:通过蛋白质组学方法,以人乳腺癌MDA-MB-435细胞为对象,筛查红景天苷干预所致的差异表达蛋白质,为后续研究红景天苷治疗乳腺癌的分子机制提供初步依据。方法:用双向电泳、软件分析和质谱分析等常用蛋白质组学技术检测4 mg/m L的红景天苷对MDA-MB-435细胞蛋白质表达谱的影响。结果:获得了红景天苷作用MDA-MB-435细胞的双向电泳图谱,初步鉴定了6个与红景天苷作用相关的差异表达蛋白质,包括细胞质肌动蛋白1、膜联蛋白A5、原肌球蛋白3亚型CRA_c、原肌球蛋白α-4链亚型2、原肌球蛋白3亚型CRA_e、酪氨酸3-单加氧酶/色氨酸。结论:成功建立了红景天苷作用于MDA-MB-435细胞的蛋白质组双向电泳分离体系,筛选出6个与红景天苷作用相关的差异表达蛋白质。
Objective: To investigate the differentially expressed proteins after treatment of human breast cancer MDA-MB-435 cells with Salidroside, which may help us to elucidate the molecular mechanism of Salidroside in the treatment of human breast cancer. Methods: The proteomics technologies such as 2-DE, software analysis, and MS were used to investigate 2-DE profiling on MDA-MB-435 cells treated with 4 mg/mL Salidroside. Results: 2-DE profilings in MDA-MB-435 cells treated with Salidroside were established, and six differentially expressed proteins were identified as actin, cytoplasmic 1,annexin AS,tropomyosin 3 isoform CRA_c,tropomyosin alpha-4 chain isoform 2,tropomyosin 3 isoform CRA_e, and tyrosine 3-monooxygenase/ tryptophan. Conclusion: 2-DE separation system for proteomes from MDA-MB-435 cells treated with Salidroside has been successfully established, and six differentially expressed proteins were identified.
