脾虚模型大鼠能量代谢相关性基因研究 被引量:12

Research on genetics correlated with energy metabolism in rats with syndrome of spleen deficiency

摘要 目的:研究脾气虚与脾阳虚大鼠模型能量代谢相关性基因的差异表达,探讨脾气虚与脾阳虚中医证候的生物学基础。方法:将24只大鼠随机分为正常组(8只)、脾气虚组(8只)、脾阳虚组(8只),采用PCR array法测定脾气虚与脾阳虚模型小肠组织能量代谢相关基因的表达水平。结果:脾气虚、脾阳虚模型共性表达下调的能量代谢相关基因包括:神经肽Y、生长抑素、血管活性肠肽;生长抑素在脾气虚模型下调表达尤为显著;神经肽Y在脾阳虚模型下调表达尤为显著。结论:脾气虚与脾阳虚模型能量代谢相关基因的表达与中医脾虚证候密切相关,为进一步阐释脾气虚与脾阳虚模型中医证候的生物学基础提供研究思路。 Objective: To study the differential expression of genetics correlated with energy metabolism between rats with syndrome of deficiency of spleen qi and syndrome of deficiency of spleen yang, and to discuss the biological basis of syndrome of deficiency of spleen qi and syndrome of deficiency of spleen yang. Methods: Twenty-four rats are randomly divided into control group, syndrome of deficiency of spleen qi group, syndrome of deficiency of spleen yang group, and with 8 rats in each group. Expression levels of genetics correlated with energy metabolism in small intestine tissue of rats with syndrome of deficiency of spleen qi and syndrome of deficiency of spleen yang are measured by using PCR array method. Results: The expressions of genetics correlated with energy metabolism are down-regulated in rats with the both syndromes include neuropeptide Y (NPY), somatostain (SST) and vasoactive intestinal peptide (VIP). The down-regulated expression of SST in rats with syndrome of deficiency of spleen qi is particularly significant, and the down-regulated expression of NPY in rats with syndrome of deficiency of spleen yang is particularly significant. Conclusion: The expression of genetics correlated with energy metabolism in rat model of syndrome of deficiency of spleen qi and syndrome of deficiency of spleen yang are closely related to the syndromes of spleen deficiency of traditional Chinese medicine, which provides a new research approach for explanation of the biological foundation of syndrome of deficiency of spleen qi and syndrome of deficiency of spleen yang.
