不同溶剂提取豆渣蛋白工艺的优化

摘要 以豆渣再利用为目的,对豆渣基本营养成分进行了含量测定。采用Tris-HCl和水两种不同溶剂对豆渣蛋白进行提取,比较并探究了豆渣蛋白提取的最佳工艺条件。结果表明,豆渣蛋白的最佳提取工艺条件为料液比1∶30(g/m L),碱提p H 10.0,碱提时间2.5 h,碱提温度40℃,酸沉p H 4.4,在此条件下豆渣蛋白得率57.36%,所得蛋白纯度48.33%。水溶剂提取蛋白得率54.24%,Tris-HCl溶剂提取豆渣蛋白的提取率较水溶剂提取高3.12%。影响豆渣蛋白提取率的先后顺序为:碱提p H>料液比>碱提时间>碱提温度。 In order to reuse soybean dregs, the basic nutrient content of soybean protein have been determined. Two different protein extraction solvents were used and the extraction condition of soybean dregs protein was optimized. It showed that Tris-HC1 solvent was higher than water solvent at protein extraction rate of soybean dregs. The optimal extraction condition was as follows: solid-liquid ratio 1 : 30 (g/mL), pH 10.0, alkaline extraction time 2.5 h, alkaline extraction temperature 40 ~C, acid precipitation 4.4. The soybean dregs protein yield was 57.36%, and the protein purity was 48.33% under this condition. Water solvent extraction yield of protein was 54.24%, and it was lower than Tris-HC1 solvent about 3.12%. The order of protein extraction rates of soybean dregs was that alkaline extraction pH〉solid-liquid ratio〉alkali extraction time〉alkali extraction temperature.
