摘要
目的:探讨黄芪注射液对大鼠高正加速度(+10Gz)暴露后嚼肌细胞凋亡的影响。方法:30只SD大鼠(雄性,200g)适应性喂养1周后,称重,随机分成5组,分别是:对照组,+10Gz组,黄芪低剂量组,黄芪中剂量组,黄芪高剂量组,每组6只,分笼饲养并做标记。各组动物每天上午腹腔注射给药1次,连续14d,给药体积为10ml/kg,低浓度组,中浓度组,高浓度组分别浓度是0.5g/ml,1g/ml,2g/ml,对照组、+10Gz暴露组给予等量的生理盐水注射。第15天进行+10Gz暴露,观察各组动物嚼肌细胞凋亡的变化。结果:黄芪中(P<0.05)、高浓度组(P<0.01)与+10Gz组相比阳性凋亡细胞明显减少,染色变浅,+10Gz暴露组与对照组相比,咀嚼肌凋亡阳性细胞数增多明显(P<0.01),高浓度组与对照组相比无显著性差异。结论:黄芪对+10Gz暴露后大鼠嚼肌细胞凋亡有明显的抑制作用。
Objective: To study the effect of astragalus membranaceu on apoptosis of masseter muscles cell in rat associated with +10Gz. Methods: 30 healthy male SD rats were randomly divided into control group, +10Gz group and low,medium, high dose of AM groups(6 rats per group). The low, medium and high doses of AM groups were fed with Astragalus membranaceu with the dosage of 0.5g/ml, 1g/ml and 2g/ml once a day for 14 days. The control group and +10Gz group were given the same amount of normal saline for 14 days. The rats were exposed to +10Gz in the 15 th day for 5 minutes, respectively. Apoptosis indexes of masseter muscles cell were observed by TUNEL. Results: Positive apoptotic masseter muscles cells in medium and high dose of AM groups decreased significantly, dyed lighter than +10Gz group(P〈0.05).+10Gz group compared with control group, the number of positive apoptotic cells increased significantly( P〈0.01).Compared with the control group, the positive apoptotic cells of high dose of AM groups did not significantly rise(P〉0.05).Conclusion: Astragalus membranaceu(AM) has significantly protective effect on the apoptosis of masseter muscles.
