葡萄膜恶性黑色素瘤中血管内皮生长因子水平与肿瘤转移的关系 被引量:4

摘要 目的:分析葡萄膜恶性黑色素瘤患者血管内皮细胞生长因子(vascular endothelial growth factor,VEGF)的表达水平是否与葡萄膜恶性黑色素瘤的转移和预后有关。方法:采用ELISA法检测葡萄膜恶性黑色素瘤患者和正常对照组中外周血VEGF水平,并采用RT-PCR和Westernblot检测葡萄膜恶性黑色素瘤患者肿瘤组织中VEGF mRNA和VEGF蛋白表达水平。对葡萄膜恶性黑色素瘤患者进行随访,用Kaplan-Meier法分析基线VEGF水平与患者术后转移的关系。结果:葡萄膜恶性黑色素瘤患者外周血VEGF水平较健康者升高。就诊时肿瘤发生转移的患者VEGF mRNA和VEGF蛋白表达水平均较未发生转移的患者升高。在随访过程中,血清VEGF≥700pg/m L的患者,肿瘤发生转移的风险较高。结论:葡萄膜恶性黑色素瘤患者中VEGF的表达水平与葡萄膜恶性黑色素瘤的转移密切相关。 AIM: To identify relationship between the expression of vascular endothelial growth factor ( VEGF) of uveal melanoma patients whether with uveal melanoma metastasis and prognosis. METHODS: The peripheral blood VEGF of uveal melanoma patients and patients in normal control group were detected by enzyme-linked immunosorbent assay (ELISA). The mRNA and protein expression level of VEGF in uveal melanoma tissues were analyzed through reverse transcription - polymerase chain reaction ( RT - PCR) and Western-blot. Uveal melanoma patients were followed up, and the relationship of baseline serum VEGF and postoperative metastasis was analyzed by Kaplan- Meier method. RESULTS: The expression level of peripheral blood VEGF was significantly increased in uveal melanoma patients than that in healthy people. The mRNA and protein expression level of VEGF in the uveal melanoma patients with metastasis when seeing a doctor were higher than patients without metastasis. In the process of follow-up, the uveal melanoma patients (serum VEGF ≥ 700pg/mL) had higher risk for tumor metastasis. CONCLUSION: The expression level of VEGF of uveal melanoma patients is closely related to uveal melanoma metastasis.
