中药复方稳心颗粒对家兔心肌缺血/再灌注损伤的影响 被引量:4

Cardioprotective effects of wenxin granules on myocardial ischemia /reperfusion injury in rabbits

摘要 目的研究复方中药制剂稳心颗粒对心肌缺血/再灌注损伤家兔模型的保护作用。方法 36只家兔随机分为假手术组(阴性对照组)、模型组、三七颗粒组(阳性对照组)及稳心颗粒低、中、高剂量组(给药组)。阳性对照组家兔灌胃三七颗粒0.4 g/(kg·d),给药组家兔分别灌胃稳心颗粒0.4、0.6、0.8 g/(kg·d),其余两组灌胃等体积生理盐水;给药1周后,采用结扎冠状动脉前降支方法制备家兔心肌缺血/再灌注模型,观察各组家兔心电图,并检测血清丙二醛(MDA)含量,以及乳酸脱氢酶(LDH)、磷酸肌酸激酶(CK)、超氧化物歧化酶(SOD)的活性。结果与假手术组比较,模型组室性心律失常发生率、血清CK与LDH活性、MDA含量升高(P<0.05),而SOD活性降低(P<0.05),表明家兔心肌缺血/再灌注模型成功;与模型组比较,阳性对照组和给药组室性心律失常发生率、血清CK与LDH活性、MDA含量均显著降低(P<0.05),SOD活性升高(P<0.05),且给药组表现出明显的剂量效应。结论稳心颗粒对家兔心肌缺血/再灌损伤有保护作用。 Objective To observe the protective effect of wenxin granules (WXG) on myocardial ischemia/ reperfusion injury in rabbits. Methods Thirty-six rabbits were randomized into 6 groups: sham operation group (negative control group), model group, Radix notoginseng group (positive control group), WXG low, middle and high dosage group (medication groups). Rabbits in above groups were given i. g. physiological saline, Radix notoginseng granule and low, middle, high dosages of WXG ( 5 mL/kg) for 7 d respectively. The myocardial ischemia/reperfusion mod- els of rabbits were induced by coronary artery occulsion. The ventricular tachycardia was recorded, the levels of CK, LDH,MDA, SOD were measured.Results Compared with sham operation group, the arrhythmia incidence, CK, LDH, MDA levels of model group increased, and the SOD activity decreased significantly ( P 〈 0. 05 ). Compared with model group, the arrhythmia incidence, CK, LDH, MDA levels of positive control group and WXG groups decreased, and the SOD activity increased significantly ( P 〈 0.05 ). In addition, there was a dose-dependent effect in WXG low, middle and high dosage groups. Contusion WXG is effective in protecting the myocardium against myocardial ischemia/reperfusion injury in rabbits.
