摘要
目的观察调节性T细胞和叉状头/翅膀状螺旋转录因子(Foxp3)在胃癌患者中的表达变化。方法随机选取2011年3月至2014年10月我院收治的胃癌患者80例为实验组,另选取同期来我院体检的80例健康人为对照组,应用流式细胞术对两组外周血中CD4+CD25+Foxp3+T细胞的表达水平进行检测,运用免疫组织化学法对胃癌及癌旁组织中的Foxp3+T细胞的表达水平进行检测,然后对两组患者的外周血中CD4+CD25+Foxp3+T细胞的表达水平、胃癌组织中的Foxp3+T细胞的表达水平、不同TNM分期、淋巴结转移患者外周血中CD4+CD25+Foxp3+T细胞的表达水平进行统计分析。结果实验组和对照组患者外周血中CD4‘CD25+Foxp3+T细胞比例分别为(6.99±1.24)%和(3.87±0.94)%,实验组显著高于对照组(P〈0.05);实验组患者胃癌组织和癌旁组织中的Foxp3+T细胞的表达水平分别为(71.44±15.64)/mm2和(21.56±9.43)/mm2,胃癌组织显著高于癌旁组织(P〈0.05);不同TNM分期、淋巴结转移患者外周血CD4+CD25+Foxp3+T细胞的表达水平之间的差异均有统计学意义(P〈0.05),不同TNM分期、分化程度、淋巴结转移患者胃癌组织中Foxp3+T细胞的表达水平之间的差异均有统计学意义(P〈0.05);外周血CD4+CD25+Foxp3+T细胞和胃癌组织Foxp3+T细胞的表达呈显著正相关(r=0.786,P〈O.05)。结论调节性T细胞和Foxp3水平在胃癌患者体内的表达提高。
Objective To analyze the expression changes of regulatory T cells and forkhead/ winged helix transcription factor P3 ( Foxp3 ) levels in patients with gastric cancer. Methods Eighty pa- tients with gastric cancer in our hospital from March 2011 to October 2014 were selected as the study group, and the 80 healthy controls for medical examination in the same period in our hospital were selected as the control group. The expression levels of CD4+ CD25+ Foxp3 + T cells in the peripheral blood were detected by flow cytometry. Immunohistochemistry was used to detect the expression of Foxp3+ T ceils in gastric carcinoma and adjacent gastric tissues. The expression of CD4+ CD25+ Foxp3 ~ T cells, expression of Foxp3 + T cells in gastric cancer, TNM stage, CD4 + CD25 + Foxp3+ T cells in peripheral blood of pa- tients with lymph node metastasis were statistically analyzed. Results The proportion of CD4 + CD25 + Foxp3+T cells in peripheral blood of the study group and the control group was ( 6. 99± 1.24 ) % and ( 3.87 ±0. 94) % ( P 〈 0. 05 ). The expression levels of Foxp3+ T in the gastric cancer tissues and adja- cent tissues in study group were (71.44 ± 15 64)/mm2 and (21.56 ±9 43)/mm2 respectiely (P 〈0. 05). The expression of CD4+ CD25+ Foxp3+ T cells in peripheral blood was related with TNM stage and lymph node metastasis (P 〈 0 05 ). The expression of Foxp3 + T cells in gastric cancer tissues was related with TNM stage, differentiation, and lymph node metastasis (P 〈 0.05 ) There was a positive correlation be- tween the expression of CD4+ CD25+ Foxp3 + T cells in peripheral blood and that of Foxp3+ T ceils in gas- tric carcinoma (r = 0. 786,P 〈 0 05). Conclusion Regulatory T cells and Foxp3 expression levels in gas- tric cancer patients are up -regulated.
