摘要
建立一种利用高效液相色谱测定白茶中8种有机酸的方法,最佳色谱条件为:Agilent-1200高效液相色谱仪,ZORBAX SB-C18色谱柱(5μm,4.6 mm×250 mm),流动相为3%甲醇-0.05 mol/L KH2PO4(用磷酸调p H至2.75),流速0.5 m L/min,柱温25℃,检测波长215 nm,进样量15μL。该方法有机酸添加回收率达到91.63%~103.34%,相对标准差为0.45%~2.34%,有机酸线性相关系数r〉0.9997,表明该测定方法快速简便,具有较高的准确度和精确度。
This paper aimed to determine how to test 8 organic acids in white tea by using high performance liquid chromatography(HPLC). The best chromatographic conditions are as follows: Agilent-1200 HPLC equipped with UV detector and a ZORBAX SB-C18column(5 μm,4.6 mm×250 mm). The mobile phase was 3% methanol-0.05 mol/L KH2P04(p H value adjusted with phosphoric acid to 2.75) with flow rate of 0.5 m L/min. Column temperature was at 25 ℃, the detection wavelength was at 215 nm and 15 μL sample was injected into the column.The recovery rate of the organic acids could reach 91.63% ~103.34% with relative standard deviations of 0.45% ~2.34%. The organic acids linear correlation coefficient was above 0.9997, indicating that the method was simple,efficient and accurate.
