HIF-1α、VEGF及CD34在大鼠骨关节炎模型软骨中的表达及意义 被引量:8

Expression and significance of HIF-1α, VEGF and CD34 in osteoarthritis articular of rat model

摘要 目的:检测低氧诱导因子(HIF-1α)、血管内皮细胞生长因子(VEGF)及CD34在膝骨关节炎(OA)软骨中的表达。方法60只Wister大鼠,鼠龄3个月,体质量约200~250 g。随机分为实验组45只和对照组15只。实验组行单膝前交叉韧带切断术,对照组不做处理,作为空白组对照;术后9周处死动物,采用免疫组织化学苏木精-伊红(HE)染色对实验组和对照组HIF-1α、VEGF、CD34表达进行检测。结果 HIF-lα、VEGF、CD34在实验组中阳性表达率分别为66.67%(30/45)、73.33%(33/45)、71.11%(32/45),均明显高于对照组;两组数据之间的差异有统计学意义(P〈0.05)。实验组HIF-1α、VEGF、CD34阳性细胞计数显著高于对照组(P〈0.05);HIF-1α与VEGF、CD34表达存在正相关(r=0.249、0.289,P〈0.05)。结论 OA软骨中HIF-1α、VEGF及CD34高表达,HIF-1α与VEGF、CD34表达存在正相关,且HIF-1α、VEGF与CD34在OA软骨退行性变进展中共同发挥着重要作用。 Objective To observe the expression and significance of hypoxia inducible factor-1α(HIF-1α), vascular endothelial growth factor(VEGF) and CD34 in osteoarthritis articular of rat model. Methods Sixty 3-month-old Wister rats with body weight 200-250 g were chosen and randomly divided into experimental group(n=45) and control group(n=15). Rats in experimental group were performed single knee transaction of anterior cruciate ligament, while no treatment was performed in control group. Rats in experimental group and control group were sacrificed 9 weeks after operation, and the expressions of HIF-1α, VEGF and CD34 were detected by immunohistochemical hematoxylin-eosin(HE) staining. Results Positive expression rates of HIF-1α, VEGF and CD34 in experimental group was 66.67%(30/45), 73.33%(33/45) and 71.11%(32/45), respectively, which were significantly higher than those of control group, and there was statistically significant difference between 2 groups (P〈0.05);Positive expression numbers of HIF-1α, VEGF and CD34 in experimental group were significantly higher than those of control group(P〈0.05). It was showed positive correlation between HIF-1αand VEGF, CD34(r=0.249, 0.289, P〈0.05). Conclusion It is demonstrated that of HIF-1α, VEGF and CD34 have high expression in rat model of osteoarthritis articular, and positive correlation exists between HIF-1αand VEGF, CD34, and they play important roles together in osteoarthritis carilage degeneration.
