摘要
目的:观察4种中药复方对糖尿病大鼠肾小球糖蛋白沉积影响。方法:取链脲霉素诱导的SD大鼠DN模型72只,分成6组,模型对照组、益气养阴方(参芪地黄汤20 g·kg-1)组、滋补肝肾方(杞菊地黄汤20 g·kg-1)组、补气养血方(当归补血汤20 g·kg-1)组、温肾健脾方(附子理中丸20 g·kg-1)组和阳性对照(缬沙坦25 mg·kg-1)组,每组12只;另取12只大鼠为正常对照组。连续经口灌胃给药8周后,进行肾脏组织HE、PAS染色病理切片和电镜肾脏病理形态学分析。结果:各给药组大鼠肾脏重量、肾重/体质量比值均有一定的降低趋势(P>0.05),肾小球截面积和肾小球体积均明显降低(P<0.05,P<0.01),滋补肝肾组、补气养血组和温肾健脾组大鼠肾小球囊腔面积显著降低(P<0.05,P<0.01),各组肾小球囊腔面积/肾小球截面积比值均有降低的趋势(P>0.05),肾小球PAS阳性面积和总光密度均显著降低(P<0.01),肾小球基底膜厚度与模型对照组比有所减轻,电子致密物沉积和足细胞足突融合、变平的现象与模型对照组比有所改善。结论:4种中药复方均能抑制糖尿病大鼠肾小球糖蛋白的沉积,4种中药复方之间比较,当归补血汤对抑制肾小球糖蛋白沉积的效果较佳。
Objective:To observe the impact of four Chinese herbal compounds on glomerular deposition glycoprotein and organic uhrastructure of diabetic nephropathy in rats. Methods : Take seventy - two STZ - induced diabetic nephropathy rats, and divide into model control group, supplementing Qi and nourishing yin group ( Shenqidihuang soup 20 g ·kg- 1 ), nourishing liver and kidney group (Qijudihuang soup 20 g · kg-1), supplementing Qi and nourishing blood group (Dang- guibuxue soup 20 g · kg- 1 ) , warming kidney and invigorating spleen group ( Fuzilizhong Pill 20 g · kg- 1 ) and positive control group (valsartan 25 mg · kg-1). Each group had 12 rats, taking another 12 normal rats for the normal control group. Administration drug for eight weeks, HE, PAS staining and electron microscope for the kidney tissue slices, and ob- serve kidney pathological morphology. Results : Each groups had certain reducing trend in the rat kidney weight, kidney weight/body weight ratio ( P 〉 0.05 ) , and significantly lower glomerulus sectional area and volume ( P 〈 0.05, P 〈 0. 01 ). Nourishing liver and kidney group ,supplementing Qi and nourishing blood group and warming kidney and invigor- ating spleen group can significantly reduce bowman capsule area (P 〈 0.05 ,P 〈 0.01 ). Each groups had reducing trend of area ratio of bowman capsule and glomerulus (P 〉 0.05), and significantly reduced glomerular PAS positive area and the total optical density( P 〈 0.01 ), moreover all can alleviate glomerular basement membrane thickness ,improve electron dense deposit and the phenomenon of podocyte foot process fusion and flatten comparing with model control group. Conclu- sion:Four kinds of Chinese herbal compounds can be in different degrees of inhibition of glomerulus glycoprotein deposi- tion, glomerular area, glomerular volume, area ratio of bowman capsule and glomerulus, improve glomerular podocyte fu-sion and flatten phenomenon and reduce the degree of thickening of basement membrane in diabetic rats.
