摘要
[目的]建立湖北利川黄连的高效液相色谱指纹图谱,为评价其质量提供科学、有效的方法。[方法]采用C18色谱柱,乙腈-0.05mol/L磷酸梯度洗脱;流速为1.0 ml/min,检测波长354 nm;柱温为35℃。[结果]对湖北利川黄连10批次药材进行指纹图谱分析,通过HPLC技术指认指纹图谱共有12个共有峰,共有峰总面积92.31%~95.67%,相似度0.940~0.986。乙腈-0.05 mol/L磷酸进行梯度洗脱最佳。[结论]该方法具有较好的稳定性和准确性,10个批次的黄连样品生成的指纹图谱可作为湖北利川黄连的标准指纹图谱。
[Objective] To establish high efficient HPLC fingerprints of Coptis chinensis from Lichuan County Hubei Province,provide scientific and efficient method for evaluating its quality. [Method] C18 chromatographic column,mobile phase of Acetonitrile- 0. 05 mol / L phosphate,flow rate at 1. 0 ml / min,gradient elution,detection wavelength at 354 nm and 35℃. The fingerprints of 10 batches samples Coptis chinensis from Lichuan County Hubei Provence were analyzed and twelve 12 common peaks were identified by HPLC. The total common peak area accounted for 92. 31%- 95. 67%,the similarity of various samples fingerprints reached 0. 940- 0. 986. [Result] Acetonitrile- 0. 05 mol / L phosphate is best for gradient elution. [Conclusion]The analytical method is stable and accurate,automatically generated fingerprint of10 batches samples of Coptis chinensis could be the standard fingerprints for Coptis chinensis from Lichuan County Hubei Province.
