摘要
目的 研究非小细胞肺癌(non-small cell lung cancer,NSCLC)患者外周血中辅助性T细胞17(T help cell 17,Th17)与调节性T淋巴细胞(T regular cell,Treg)表达及平衡关系的变化,探讨其在NSCLC发病机制中的作用.方法 应用流式细胞术检测60例NSCLC患者和31例健康对照外周血Th17和Treg的表达及平衡关系的变化.结果 NSCLC患者外周血Th17表达明显高于健康对照组[(3.40%±0.49%) vs.(2.16%±0.39%),P<0.001],但腺癌与鳞癌组及进展期(Ⅳ期)与局限期(Ⅰ~Ⅲ期)组Th17无明显差异.NSCLC患者外周血Treg表达与健康对照相比也有显著升高[(7.76%±1.58%) vs.(4.40%±1.08%),P<0.001],并且进展期较局限期组Treg表达明显升高[(8.16%±1.56%) vs.(6.97%±1.35%),P=0.005].腺癌组与鳞癌组Treg表达无明显差异.NSCLC组Treg/Th17较健康对照组明显增高[(2.36±0.73) vs.(2.05±0.46),P=0.038],并且进展期较局限期组进一步升高[(2.53±0.78) vs.(2.03±0.51),P=0.012].结论 NSCLC患者可能存在Treg/Th17的失衡,而Treg/Th17比值的变化也与NSCLC的预后相关.
Objective To observe the changes and imbalance of T help cell 17 (Th17) and T regular cell (Treg) in peripheral blood of patients with non-small cell lung cancer (NSCLC).Methods Peripheral bloods were collected from 60 patients with NSCLC and 31 healthy subjects.The percentages of Th17 and Treg in peripheral blood were detected by flow cytometry.Results The percentage of Th17 in peripheral blood was significantly increased in patients with NSCLC compared with the controls [(3.40% ± 0.49%) vs.(2.16% ± 0.39%),P<0.001],but there was no difference in the subgroups of NSCLC no matter when classify with tumor type or tumor stage.The percentage of Treg in peripheral blood was also significantly higher in patients with NSCLC compared with the control [(7.76% ± 1.58%) vs.(4.40% ± 1.08%),P<0.001].Moreover,the patients in advanced stage had higher percentage of Treg compared with those in limited stage [(8.16% ± 1.56%) vs.(6.97% ± 1.35%),P =0.005].The ratio of Treg and Th17 (Treg/Th17) in patients with NSCLC was increased greatly compared with the controls [(2.36 ± 0.73) vs.(2.05 ± 0.46),P =0.038].Moreover,the Treg/Th17 ratio also increased in the patients in advanced stage.Conclusions The Th17/Treg imbalance might play a critical role in the pathogenesis of NSCLC,which could be an important factor correlated with the prognosis of NSCLC.
