表没食子儿茶素没食子酸酯对慢性间歇性缺氧大鼠的心肌保护作用

摘要 目的探讨表没食子儿茶素没食子酸酯(EGCG)对慢性间歇性缺氧大鼠心肌的保护作用。方法将24只SD大鼠随机分为4组,每组6只。分为常压常氧组和慢性间歇性缺氧组(A组生理盐灌胃,B组50 mg/kg EGCG灌胃,C组100 mg/kg EGCG灌胃)。分别在常压常氧和慢性间隙缺氧环境下饲养。8周处死动物,检测血清中过氧化物歧化酶(SOD)、谷胱甘肽过氧化酶(GSH-PX)活性及乳酸脱氢酶(LDH)含量。结果与慢性间歇性缺氧生理盐水组相比,EGCG组能明显抑制心脏的肥厚,并能升高SOD和GSH-PX的活性,降低LDH的含量,并呈剂量依赖性。结论 EGCG对慢性间隙缺氧性缺氧大鼠心肌具有一定的保护作用,这与其抗氧化作用有关。 Objective To explore the role of epigallocatechin-3-gallate( EGCG) in protection of cardiac muscle in rats with chronic intermittent hypoxia. Methods Twenty-four SD rats were randomly divided into four groups,6 rats in each group. The control group was fed with saline in the normal air. The experimental groups( named group A,B and C) were fed with saline,50 mg / kg EGCG and 100 mg / kg EGCG respectively in chronic intermittent hypoxia conditions. After 8 weeks,all rats were killed. The serum activity of SOD,GSH-PX and level of LDH were detected. Results Compared with the rats fed by saline in the experimental group,EGCG groups showed significant inhibitions on myocardial hypertrophy,and EGCG was able to increase the activity of SOD,GSH-PX and decrease the level of LDH in serum( P〈0. 05),and the effect was dose-dependent. Conclusion EGCG can significantly protect the heart from injury caused by chronic intermittent hypoxia in rats,and this function is correlated with its antioxygenation.
