摘要
以毕赤酵母重组菌(Pichia pastoris)GS115-Ch-Glu为研究对象,进行了亚麻子发酵脱毒工艺的研究,并对重组菌的发酵培养条件进行优化。结果表明,毕赤酵母重组菌GS115-Ch-Glu发酵扩大培养的培养基最佳碳源为2.5%玉米淀粉,最佳氮源为5.0%黄豆粕。优化后的基础发酵培养基发酵条件为温度28℃、转速250 r/min、发酵时间33 h、初始p H 6.5、接菌量1.5%、装液量50 m L/250 m L。在此最佳发酵条件下,菌体产量为9.0×1010个/m L。
Pichia pastoris GS115-Ch-Glu as a sort of useful recombinant yeast was used to detoxify in linseed. Its fermentation conditions were optimized. The results showed that the optimal carbon and nitrogen source were 2.5% corn starch and 5.0% soybean meal. The optimal fermentation conditions in basal medium were the temperature of 28 ℃, the speed of 250 r/min, the incubation time of 33 hours, the initial pH value of 6.5, the inoculation rate of 1.5% and the liquid volume of 50 mL/250 mL. Under these conditions, the fermentation yield of the cell production was per milliliter of 9.0× 10^10 strains.
