HPLC Determination of Six Ingredients in Ramulus Cinnamomi and Poria Cocos Extract

摘要 目的:建立高效液相色谱法测定桂枝、茯苓药对提取物中原儿茶酸、香豆素、桂皮醇、肉桂酸、桂皮醛、茯苓酸6个成分的含量。方法:采用Diamonsil C18色谱柱(200 mm×4.6 mm,5μm),流动相乙腈-0.05%磷酸水,梯度洗脱,流速1.0 m L·min-1,检测波长254 nm,柱温25℃,对药对桂枝中5个成分同时测定;以乙腈-0.05%磷酸水(75∶25)为流动相,流速1.0 m L·min-1,检测波长210 nm,柱温25℃,对药对茯苓中茯苓酸的含量测定。结果:原儿茶酸、香豆素、桂皮醇、肉桂酸、桂皮醛和茯苓酸分别在0.00818~0.0818μg(r=0.9995),0.03866~0.3866μg(r=0.9999),0.03138~0.3138μg(r=0.9998),0.03512~0.3512μg(r=0.9998),0.3522~3.522μg(r=0.9998),0.03920~0.3920μg(r=0.9998)范围内有良好的线性关系。原儿茶酸、香豆素、桂皮醇、肉桂酸、桂皮醛和茯苓酸的加样回收率(n=6)分别为99.2%,98.9%,100.0%,98.4%,99.2%,98.9%,RSD均〈3%。结论:该方法简便、准确,可测定桂枝、茯苓药对提取物中6个成分的含量,为其质量控制提供参考。 Objective:To establish an HPLC method for determination of six ingredients protocatechuic acid,coumarin,cinnamic alcohol,cinnamic acid,cinnamaldehyde,pachymic acid in Ramulus Cinnamomi and Poria cocos extract.Methods:The sepatation was performed on a Diamonsil C18column(200 mm × 4.6 mm,5 μm) with the gradient elution of acetonitrile and 0.05% aqueous phosphoric acid at the flow rate of 1.0 m L·min-1.Detection wavelength was set at 254 nm and the column temperature was maintained at 25 ℃ for determination of above-mentioned five ingredients in Ramulus Cinnamomi.And the mobile phase was acetonitrile-0.05% aqueous phosphoric acid(75∶ 25) at the flow rate of 1.0 m L·min-1.Detection wavelength was set at 210 nm and the column temperature was maintained at 25 ℃ for determination of above-mentioned pachymic acid in Paria cocos.Results:Protocatechuic acid,coumarin,cinnamic alcohol,cinnamic acid,cinnamaldehyde and pachymic acid had good linear relationships in the ranges of 0.00818 ~ 0.0818 μg(r = 0.9995),0.03866 ~ 0.3866 μg(r = 0.9999),0.03138 ~ 0.3138 μg(r= 0.9998),0.03512 ~ 0.3512 μg(r = 0.9998),0.3522 ~ 3.522 μg(r = 0.9998),0.03920 ~ 0.3920 μg(r = 0.9998),respectively.The average recoveries(n = 6) were 99.2%,98.9%,100.1%,98.4%,99.2% and 98.9% respectively with RSD values of the recovery were less than 3%.Conclusion:The method is simple and accurate for the determination of six ingredients in Ramulus Cinnamomi and Poria cocos extract.The method is also helpful to control the quality of Ramulus Cinnamomi and Poria cocos extract.
