二甲基亚硝胺对不同年龄及性别小鼠急性肝损伤的影响 被引量:3

Effects of dimethylnitrosamine on acute toxic liver injury in different age and gender mice

摘要 目的:研究年龄与性别对二甲基亚硝胺(DMN)中毒所致小鼠急性肝损伤的影响。方法:采用一次性DMN 17.5μl/kg腹腔内注射染毒致4、8、12周龄雌雄性小鼠急性肝损伤,通过肝功能血清生化指标[丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)、直接胆红素(DBil)、血清白蛋白(Alb)和总胆固醇(TC)]检测和肝组织HE染色病理学检查,观察不同性别和年龄小鼠急性肝损伤变化特征。结果:DMN染毒48h后致4周龄小鼠血清ALT、AST上升和肝内出血性坏死,以雌性小鼠病变更为明显,部分雌性小鼠出现腹水。8周龄和12周龄小鼠血清ALT、AST上升和肝内出血性坏死更为明显,以雌性小鼠病变极为严重,12周龄小鼠肝呈花斑状,雌性小鼠全部出现腹水并出现死亡,8周龄和12周龄小鼠各有2只雄性小鼠出现腹水。结论:DMN中毒所致小鼠急性肝损伤与年龄和性别密切相关,以雌性、成年小鼠更加明显,表现为肝内出血性坏死和腹水为特征。 Objective:To study the effects of age and gender on dimethylnitrosamine (DMN) -induced acute toxic liver injury in mice. Methods: Male and female mice of 4,8,12 weeks old were intraperitoneally injected with DMN with dosage of 17.5μl /kg once a time to in-duce acute liver injury, after 48 hours serum biochemical parameters of liver function [including albumin(Alb), alanine aminotransferase (ALT) ,aspartate aminotransferase (AST), direct bilirubin (DBil),total cholesterol(TC)] and HE staining of liver tissue were detected to observe the characteristic changes of acute liver injury with different age and gender.Results:Forty-eight hours after the intraperitoneal injec-tion of DMN with dosage of 20mg/kg once a time caused increase of serum ALT, AST and DBil, decrease of serum of Alb and TC,hemor-rhagic necrosis of the liver in mice of 4 weeks old.Those changes were significantly evident in the female mice, some female mice had asci-tes.With the increase of age, the above changes became more evident, all female aged mice of 8 and 12 weeks old had serious ascites, and some male aged mice had ascites, deaths only occurred in female aged mice of 8 and 12 weeks old.Conclusion:The acute liver injury in mice induced by DMN is closely related with age and gender, the injury is more pronounced in female aged mice,which manifestations are hemor-rhagic liver necrosis and ascites.
