阿司匹林诱导肿瘤细胞凋亡促进钙网蛋白表达的研究 被引量:2

摘要 目的探讨阿司匹林对肿瘤细胞凋亡及肿瘤细胞钙网蛋白(calreticulin,CRT)表达的影响。方法选择人的肝癌细胞系Hep G2作为目标细胞,用不同浓度的阿司匹林培养目标细胞,光镜下观察细胞活性,采用MTT法检测肿瘤细胞的凋亡,免疫荧光方法检测肿瘤细胞钙网蛋白表达量。结果 5μmol/ml,10μmol/ml,15μmol/ml浓度下的阿司匹林培养的肿瘤细胞,其对应的肿瘤细胞的抑制率分别为17.9%±1.65%,33.25%±3.77%,61.25%±3.86%,同时,对应肿瘤细胞钙网蛋白的表达量呈递增趋势。结论阿司匹林可以抑制人肝癌细胞Hep G2的生长,促进肿瘤细胞钙网蛋白的表达量,从而为肿瘤的免疫治疗提供了一个新的思路。 Objective To disuss the impact of Aspirin on tumor cell apoptosis and tumor calreticulin,expression.Method Human hepatocellular carcinoma cell line Hep G2 as target cells were choosen,with different concentrations of aspirin training target cells,then observe the cell activity with Light microscopy,detect the apoptosis of tumor cells by MTT,immunofluorescence method was used to detect tumor cell calcium net amount of protein expression. Results Tumor cells cultured by aspirin with 5μmol/ml,10μmol/ml,15μmol/ml concentrations,and their corresponding the inhibition rate of tumor cells were 17.9%±1.65%,33.25%±3.77,61.25%±3.86,at the same time,corresponding to the expression of tumor cell calcium net protein quantity increasing trend. Conclusion Aspirin can inhibit the growth of human hepatocellular carcinoma cells Hep G2,promote the expression of tumor cell calreticulin quantity,for tumor immunotherapy provides a new train of thought.
