摘要
目的探讨替米沙坦对DOCA-盐型高血压大鼠氧化应激及血管内皮功能的影响。方法以SD大鼠复制DOCA-盐型高血压大鼠模型,设假手术组(6只),DOCA-盐型组(DOCA-模型组,6只);替米沙坦干预组(DOCA-替米沙坦组,6只),氨氯地平对照组(DOCA-氨氯地平组,6只)。造模成功后,DOCA-替米沙坦组大鼠给予灌服替米沙坦10mg/(kg·d),DOCA-氨氯地平组给予灌服氨氯地平5mg/(kg·d),假手术组和DOCA-模型组给予灌服等体积的生理盐水,干预时间为1月;每周测鼠尾血压,实验结束后测血清中SOD、MDA水平;分离肠系膜动脉观察各组大鼠血管功能(包括血管内皮依赖性舒张功能及非内皮依赖性舒张功能)。结果替米沙坦与氨氯地平均可显著降低DOCA-盐型大鼠的血压,差异有统计学意义(P<0.05);DOCA-替米沙坦组大鼠血浆MDA水平显著低于DOCA-模型组及DOCA-氨氯地平组,SOD水平显著高于DOCA-模型组及DOCA-氨氯地平组,差异有统计学意义(P<0.05);替米沙坦可显著改善DOCA-盐型大鼠血管内皮依赖性舒张功能,差异有统计学意义(P<0.05);而DOCA-氨氯地平组与DOCA-模型组比较,差异无统计学意义(P>0.05)。结论替米沙坦不但可显著降低DOCA-盐型大鼠的血压,还可显著降低其氧化应激水平,改善血管内皮功能,但相关机制有待进一步探究。
Objective To explore the effects of telmisartan on oxidative stress and endothelial function in DOCA-salt hypertension rats. Methods Sprague Dawley rats were used to establish DOCA-salt hypertension model. Rats were divided into Sham group ( n = 6 ), DOCA-salt group ( n = 6 ), DOCA-salt-telmisartan group ( n = 6 ) and DOCA-salt-amlodipine group(n= 6). After the model was erected, rats in DOCA-salt-telmisartan group were given telmisartan[10 mg/(kg · d)]by gavage;rats in DOCA-salt amlodipine group were given amlodipinc[5 mg/(kg ·d)] by gavage, rats in sham group and DOCA-salt group were given the same volume of saline. Tail blood pressure was measured in each week,after four weeks the rats were sacrificed,and the SOD, MDA levels in serum were measured by kits;endothelium dependent relaxation induced by acetylcholine and endothelium-independent relaxation induced by nitroglycerin in rats" mesenterie arteries were also measured. Results Both telmisartan and amlodipine decreased the blood pressure levels significantly in DOCA-salt rats(P〈0.05) as compared to sham group. Rats in telmisartan group has a lower MDA level hut a higher SOD level as compared to DOCA-salt or amlodipine group(P〈0.05); Four weeks of telmisartan administration improved the endothelium dependent vasorelaxation in mesenteric arteries of DOCA-salt rats(P〈0.05) as compared to DOCA-salt rats;there was no significant difference between amlodipine and DOCA salt group(P〉0.05). Conclusion Telmisartan not only reduces the DOCA-salt rats blood pressure,hut also decreases the oxidative stress levels, thus improves endothelial function, but the related mechanism remains to be further explored.
