摘要
试验旨在揭示京海黄鸡翅重性状的遗传基础,寻找可用于京海黄鸡翅重性状改良的分子标记。本研究以京海黄鸡核心群母鸡为基础,测定了屠宰时的单侧翅重,利用简化基因组测序技术进行全基因组关联研究(GWAS),检测与翅重相关的SNPs位点。结果表明:共检测到7个与翅重相关的SNPs位点,其中,rs2011502599、rsz26128672、rs1830645和rs475641139 4个SNP位点达到全基因组显著水平(P〈5.5E-07),rs476249085、rs475679707和rs473712263 3个SNP位点达到全基因组潜在显著水平(P〈1.1E-05);筛选每个显著SNP周围1Mb区域内的基因,共找到7个可能的候选基因,分别为PGO2、SMARCA2、ZNF302、QDPR、FBXL5、LDB2、PPARGCIA 7个基因;使用G0数据库对7个候选基因的细胞组分、分子功能和参与的生物进程进行分析发现,4个SNPs集中分布在4号染色体上的73.71~76.25Mb的区域内。表明PGO2、SMARCA2、ZNF302、QDPR、FBXL5、LDB2、PPARGC1A 7个基因和4号染色体73.71~76.25Mb区域可能为影响京海黄鸡翅重性状的重要候选基因和区域。
In order to reveal the genetic basis of wing weight trait and scan molecular markers which can be used to improve this trait, wing weight of Jinghai yellow chicken in core group was measured after being slaughtered. The SNPs were detected using SLAF-seq technology. Finally, we got 7 SNPs that associated with wing weight. Rs2011502599, rsz26128672, rs1830645 and rs475641139 reached 5% Bonferroni genome-wide significance (P〈 5. 5E-07) and rs476249085, rs475679707 and rs473712263 reached "suggestive" genome-wide significance (P〈 1.1E-05). Scan the 1Mb region (candidate SNP position+0.5 Mb) and seven candidate genes (PG02, SMARCA2, ZNF302, QDPR, FBXL5, LDB2, PPARGC1A) were obtained. The cellular component, molecular function and biological process were analyzed by using GO database. Furthermore, four SNPs were distributed in the region of 73.71-76.25Mb on chromosome 4. conclusion, PG02, SMARCA2, ZNF302, QDPR, FBXL5, LDB2, PPARGC1A and 73.71-76.25 Mb region on chromosome 4 were important candidate genes and region affecting wing weight trait in Jinghai yellow chcken.
