(±)-2-氨基-3-亚甲基戊酸盐酸盐的全合成

Total Synthesis of(±)-2-Amino-3-methylene Pentanoic Acid Hydrochloride

摘要 (±)-2-氨基-3-亚甲基戊酸盐酸盐是一种具有生物活性的天然产物,该文以丝氨酸为原料,经过Grignard反应、氧化反应、Wittig反应、Jones氧化等单元反应,合成(±)-2-氨基-3-亚甲基戊酸盐酸盐,总收率为39%。考察并优化了氧化反应、Grignard反应和Wittig反应的工艺条件。该合成方法也适用于β取代的β,γ-不饱和α-氨基酸的合成。 ( ±)-2-Amino-3-methylene pentanoic acid hydrochloride is a biologically active natural product,which was synthesized via a series of elementary reaction,including Grignard reaction,oxidation,and Wittig reaction,using serine as the raw material. Finally,the target compound was obtained in ten steps with an overall yield of 39%. The reaction conditions of oxidation,Grignard reaction and Wittig reaction were optimized. The improved process can be applied to the synthesis of β-substituted vinylglycine analogues.
