摘要
以甜菜制糖废弃物甜菜粕为原料,采用钙离子螯合剂乙二胺四乙酸二钠(EDTA)提取甜菜果胶,并对其提取工艺进行优化。在单因素试验的基础之上,对影响果胶得率的主要因素提取温度、提取时间、液固比及EDTA浓度,进行Box-Benhnken中心组合试验设计,以果胶得率为响应值,进行响应面分析;同时建立EDTA提取甜菜果胶的二次回归模型。试验结果表明,建立的数学模型能很好地反映各因素与果胶产率之间的关系;甜菜果胶的最佳提取工艺为提取时间2.29 h,液固比24.6∶1(m L/g),螯合剂EDTA浓度1.0%,提取温度90℃,此时果胶得率为21.91%,与模型预测值接近。同时,采用间羟联苯法测定了甜菜果胶的半乳糖醛酸含量为71.26%;红外光谱的数据表明,螯合剂法提取的果胶样品具有果胶分子的结构特征。
Sugar beet pulp, a by-product of sugar industry, was investigated as a source of pectin. The pectin was extracted with calcium chelating agent disodium ethylenediaminetetraacetate (EDTA). Extraction temperature, extraction time, ratio of liquid to solid and concentration of EDTA affecting the yield of pectin were optimized by using response surface methodology (RSM) on the basis of single-factor tests. The results showed that the generated regression model adequately explained the data variation and significantly represented the actual relationship between the independent variables and responses. The optimum exlracfion time, ratio of liquid to solid, concentration of EDTA and extraction temperature were 2.29 h, 24.6 : 1 (mL/g), 1.0%, 90 %. Under these conditions, the experimental extraction yield of sugar beet pectin was 21.91%, which was well in agreement with the predicted one. The total galacturonic acid content was 71.26%, which was determined with metahydroxyldiphenyl method. FT-IR results indicated that the obtained sugar beet pectin was with the character of molecular structure of pectin.
