摘要
目的研究中药何首乌主要有效成分二苯乙烯苷(THSG)对多柔比星(DOX)致心肌细胞凋亡的保护作用及机制。方法分离培养乳鼠心肌细胞,观察DOX单独及与THSG联合处理对心肌细胞的凋亡、线粒体膜电位及凋亡相关蛋白表达的影响。结果 THSG能够剂量依赖性地减少DOX诱导的心肌细胞凋亡、线粒体膜电位下降、抑制caspase-3激活、增加Bcl-2及减少Bax的蛋白表达。结论 THSG对DOX诱导的心肌细胞凋亡具有保护作用,且作用与其能够减少线粒体膜电位下降、抑制caspase-3激活,增加Bcl-2及减少Bax的蛋白表达有关。
Objective To investigate the protective effect and mechanism of 2,3,5,4'-tetrahydroxystilbene-2-O-β-D-glucoside(THSG)on cardiomyocyte apoptosis induced by doxorubicin(DOX).Methods The neonatal rats' cardiomyocytes were isolated and cultured.The effect of DOX alone and DOX combined treatment with THSG on apoptosis,mitochondrial membrane potential(MMP)and apoptosisrelated proteins expression were observed.Results THSG could dose-dependently ameliorate DOX-induced apoptosis and the loss of MMP,inhibit caspase-3activation,increase the expression of Bax protein and reduce the expression of Bcl-2 protein,respectively. Conclusion THSG could ameliorate the cardiomyocyte apoptosis induced by DOX involving alleviating loss of MMP,caspase-3activation and changes of Bax protein expression.
