摘要
参照国标法测定饲用植酸酶活性,研究不同来源的底物植酸钠、底物浓度、底物pH对饲用植酸酶活性的影响。结果表明,以P8810为代表的植酸钠盐水合物作为底物,其测定出的植酸酶酶活显著高于使用以P3168为代表的植酸钠作为底物所测定的植酸酶酶活。以P8810作为底物时,其反应浓度在高于6.6 mmol/L时,会产生底物抑制效应。以P8810作为底物时,调节底物pH至国标中定义的5.5与未调节pH所测得的植酸酶酶活相比较,差异并不显著,且在低底物pH条件下,某些植酸酶同样能够释放出最大催化活性。因此现有的国标法测定饲用植酸酶活性已与实际应用有一定的差别,不能照搬国标方法测定现有饲用植酸酶产品。
The activity of feed phytase was determined with reference to the national standard method,the study discussed the influence of the substrate phytic acid sodium from different source,concentration and pH of substrate on the activity of feed phytase.The results showed that when phytic acid dodecasodium salt hydrate was represented by P8810 as substrate,the activity of phytase determined was significantly higher than sodium phytase represented by P3168 as substrate. When P8810 was used as substrate and the concentration in the reaction was higher than 6.6 mmol/L,inhibitory action of substrate would be generated.The activity of phytase had no significant difference whether adjusting pH to 5.5 which was defined in national standard,and at low pH of substrate,some phytase could also release the maximum catalytic activity. Therefore,the existing national standard method for the determination of feed phytase activity had some differences with the actual application,we could not copy the existing national standard method for the determination of feed phytase products.
