摘要
目的:研究环境雌激素(environmental estrogens,EEs)中壬基酚(NP)对体外培养子宫肌瘤细胞的作用效应及可能机制。方法:对子宫肌瘤细胞进行原代、传代培养及细胞鉴定。CCK-8法分析不同浓度NP在24小时、48小时、72小时对子宫肌瘤细胞活性的作用效应。采用荧光定量RT-PCR法检测各干预组雌激素受体α(ERα)mRNA、胰岛素样生长因子-1(IGF-1)mRNA、血管内皮生长因子(VEGF)mRNA的表达。采用Western Blot法检测ERα、IGF-1、VEGF蛋白的表达情况。结果:不同浓度的NP作用于子宫肌瘤细胞24小时、48小时、72小时,结果显示NP对子宫肌瘤细胞有促增殖作用,这种作用具有时间和浓度的依赖性。不同浓度的NP促进ERα、IGF-1、VEGF相关基因和蛋白的表达,该作用随着NP作用浓度的增加而增强,差异有统计学意义(P<0.05)。结论:NP可促进子宫肌瘤细胞增殖,其作用机制可能与通过ERα介导的IGF-1、VEGF等促肿瘤生长因子的表达增加有关。
Objective:To study the effect of environmental estrogens nonylphenol(NP)on uterine leiomyoma(UL)cells. Methods: Primary UL cells were cultured and treated with NP in various doses and durations (24,48 and 72 hrs). The viability of cells were determined by CCK-8,and the expressions of ERa,IGF-1 and VEGF were detected by real-time quantitative PCR and western blot. Results:NP promoted the growth of UL cells and the expressions of ER(x,IGF-1 and VEGF mRNA and protein. The viability of cells treated with NP was time-and dose-dependent. Conclusions: NP could promote the growth of UL cells,which might be associated with the ER-rnediated activation of growth factors such as IGF-1 and VEGF.
