缺氧对人微血管内皮细胞HIF-VEGF-Notch信号通路的影响 被引量:4

摘要 目的观察缺氧对人微血管内皮细胞(HMVEC)中HIF-VEGF-Notch信号通路相关分子表达的影响。方法常规方法培养HMVEC细胞,低氧培养箱制备HMVEC缺氧模型,正常细胞作为对照组。采用Western Blot法检测HMVEC中HIF-1α及Notch-1的蛋白表达,ELISA法检测细胞培养上清液中VEGF蛋白表达水平。结果缺氧组和对照组细胞中Notch1蛋白表达分别为2.07±0.16和0.91±0.04,两者差异有显著意义(P<0.05);HIF-1α蛋白表达缺氧组为2.53±0.19,对照组为0.67±0.08,差异有统计学意义(P<0.05)。对照组上清液中VEGF表达量为50.04±5.92 ng/L,缺氧组上清液中VEGF表达量为238.04±19.29 ng/L,两者差异有统计学意义(P<0.05)。结论缺氧可上调HMVEC细胞中HIF-VEGF-Notch信号通路相关分子的表达,此可能与重要脏器缺血缺氧性损伤后诱导的血管新生有关。 Objective To observe the effects of hypoxia on the expression of the HIF-VEGF-Notch signaling pathway-related factors in human microvascular endothelial cells (HMVEC). Methods To cultured HMVEC in the conventional condition. Putting HMVEC under hypoxia incubator to establish hypoxic model,the normal cells as a control group. The protein levels of HIF-1α and Notch-1 were detected by Western Blot,the production of VEGF protein in the cell culture supernatants were detected by ELISA. Results The expression of Notchl protein in hypoxia group and control group cells were 2.07±0.16 and 0.91±0.04 respectively,the difference was statistically significant (P〈0.05). The protein levels of HIF-1α in hypoxia group was 2.53±0.19,control group was 0.67±0.08 (P〈0.05). The expression of VEGF protein in the supernatant of control group and hypoxic group were 50.04± 5.92ng/L and 238.04±19.29ng/L respectively (P〈0.05). Conclusion The expression of the HIF-VEGF-Notch signaling pathway- related factors in HMVEC were upregulated by hypoxia,which is related to the angiogenesis in the important organs after ischemi- a-hypoxia injury.
