摘要
旨在筛选出分离纯化发酵液中米尔贝霉素A3/A4的较佳大孔吸附树脂以及工艺条件.采用静态吸附与解吸、动态吸附与解吸相结合的方法,以树脂的最大吸附量、解吸率为考察指标,确定最佳的纯化工艺.结果表明:HZ816树脂对米尔贝霉素A3/A4有较好的吸附与解吸效果,吸附等温线属Langmiur型.其较佳的工艺条件为粗提液中米尔贝霉素A3/A4的质量浓度为1 212mg/L,上样流速1.5BV/h,吸附1.5h后,先用7BV的70%乙醇溶液进行洗涤,然后用4BV的90%的乙醇溶液进行洗脱,米尔贝霉素A3/A4的质量浓度可由纯化前的6.4%提升至16.8%,解吸率大于78%.应用HZ816大孔树脂对发酵液中的米尔贝霉素A3/A4进行分离纯化,其工艺简单稳定,具有吸附率和解吸率高、洗脱剂安全低毒的特点,具有较高的应用价值.
Various macroporous resins with different properties were employed to purify milbemycin A3/A4 from the fermentation broth.Static and dynamic adsorption and desorption experiments were conducted,meanwhile,the adsorption and desorption rates of milbemycin A3/A4 were concerned to test out the prominent macroporous resin and optimal processing parameters.HZ816 was subsequently selected as the best macroporous resin,and the adsorption data are agree with the Langmiur isotherm.The optimal processing parameters were obtained as follows:the crude extract containing milbemycin A3/A4 of 1 212mg/L was loaded at a flow rate of 1.5BV/h,after adsorption of 1.5h,the HZ816 column was washed with 7BV of 70%ethanol,for removing the impurities,and eluting the aimed product with 4BV of 90%ethanol.As a result,the content of purified milbemycin A3/A4 was increased from 6.4%to 12.8%,the desorption rate was over 78%.The method is a simple,safe and efficient technology,and has a promising potential of purifying milbemycin A3/A4 from the fermentation broth.
