百草枯中毒大鼠肺组织高迁移率族蛋白B1的表达及意义 被引量:3

Expression and Significance of High Mobility Group Box 1 Protein in Lung Tissues of Paraquat Poisoned Rats

摘要 目的观察高迁移率族蛋白B1(HMGB1)在百草枯中毒致急性肺损伤(ALI)大鼠肺组织的表达情况,探讨HMGB1在百草枯中毒致ALI发病中的作用。方法大剂量百草枯一次性灌胃建立ALI大鼠动物模型,观察各组大鼠肺组织病理,采用酶联免疫吸附试验测定肺组织中HMGB1表达情况。结果正常大鼠肺组织有少量HMGB1表达。百草枯染毒后,HMGB1表达在12 h迅速升高,至染毒后24 h达最高峰,随后有所下降,12 h后大鼠肺组织HMGB1表达水平明显高于正常对照组(P<0.05)。结论正常大鼠肺组织有一定水平HMGB1的表达,百草枯染毒后,HMGB1表达异常增高,其可能促进百草枯中毒时ALI的发生与发展。 Objective To investigate the expression of high mobility group box 1 protein( HMGB1) in lung tissues of the paraquat poisoned rats with acute lung injury( ALI),and to explore the role of HMGB1 in ALI induced by paraquat. Methods The rat model of ALI was established by intragastric administration with high dosage of paraquat.Pathological alteration was examined and the expression of HMGB1 was detected by enzyme-linked immunosorbent assay( ELISA) in lung tissues of the rats in each group. Results Slight HMGB1 expression was observed in lung tissues of normal rats. The level of HMGB1 increased rapidly within 12 hours and reached the peak 24 hours after paraquat infection.Then it decreased again. Twelve hours later,HMGB1 expression in the infected group was significantly higher than that in the normal control group( P〈0. 05). Conclusion The lung tissues of normal rats can express HMGB1 to a certain extent.After paraquat infection,the expression of HMGB1 increases abnormally,which may contribute to the occurrence and development of ALI.
