摘要
目的探讨胶质细胞源性神经营养因子(GDNF)和脑源性神经营养因子(BDNF)在大鼠脑组织中的表达水平随损伤时间的变化规律,以期为脑损伤时间的推断提供新的实验依据。方法将90只无特定病原体(SPF)SD大鼠随机分为正常对照组(n=10)及实验组(n=80),对实验组大鼠建立改良的大鼠弥漫性脑损伤模型,并根据伤后时间分为1、3、6、12、24、48、72、120 h组,每组各10只。采用免疫组织化学方法检测各组大鼠脑组织中GDNF和BDNF表达,比较各组大鼠脑组织不同部位及损伤时间GDNF、BDNF的表达和变化规律。结果大脑、小脑、脑干组织中GDNF和BDNF阳性信号灰度在损伤后1 h组表达升高,6 h组达最高峰,12 h组开始回落;1、3、6、12、24、48、72、120 h组大脑、小脑、脑干组织中GDNF阳性信号灰度值均显著高于正常对照组(P<0.05),各组各部位阳性信号面积密度比较,差异无统计学意义(P>0.05)。结论 BDNF、GDNF可以作为大鼠弥漫性脑损伤后经过时间推断的实验指标。
Objective To investigate the changes of GDNF and BDNF in rats after diffuse brain injury and provide new experimental evidence for estimating time of brain injury. Methods 90 SD rats of SPF stage were divided in normal control group(n=10) and experiment group(n=80) randomly. Diffuse brain injury model was established in the experiment group, and these rats were divided into 1, 3, 6, 12, 24, 48, 72 h and 120 h group according to the injury time, 10rats in each group. The expression of GDNF and BDNF of each group was detected by immunohistochemical, the expression of GDNF and BDNF of each group in different part and injury time were compared. Results The expression of GDNF and BDNF in cerebrum, cerebellum and brain stem of the 1 h group increased, peak of the 6 h group, the 12 h group started to come down; the expression of GDNF in cerebrum, cerebellum and brain stem of 1, 3, 6, 12, 24, 48, 72 h and 120 h group were higher than those of normal control group(P〈0.05), area density of positive signal of different part in different group had no statistically significant difference(P〈0.05). Conclusion BDNF and GDNF can be used as marks to estimate the time of brain injury.
