摘要
目的 观察补肾复方对老年人 T细胞凋亡及其相关基因群 m RNA表达的调控作用模式。方法 老年人 44例 ,采取补肾方和安慰剂随机分组 ,双盲给药 ;采用 TUNEL标记的流式细胞仪检测分析各组 T细胞凋亡的百分率 ;采用荧光实时定量 PCR技术分析各组 T细胞凋亡相关促凋亡基因 (Fas、Fas L、TNFR1、Bax)及抗凋亡基因 (TNFR2、bcl- 2 )的转录情况。结果 补肾组 T细胞凋亡率比安慰剂组明显降低 (P<0 .0 1 ) ,Fas L、TNFR1基因 m RNA表达明显降低 (P<0 .0 5) ,bcl- 2基因 m RNA表达显著增高 (P<0 .0 5)。结论 补肾方对老年人 T细胞部分促凋亡基因的转录具有负调控作用 ,同时上调抗凋亡基因的转录 ,这种协同作用模式是补肾方下调老年人 T细胞过度凋亡的分子机制之一。
Objective To observe the regulating pattern of kidney tonifying recipe on expression of T lymphocytes apoptosis related genes in aging subjects. Methods A randomized double blind placebo controlled trial was used to elucidate the effect of Tonifying Kidney recipe on the peripheral T lymphocytes apoptosis percentage by method of TUNEL and flowcytometry assay, as well as the expression of pro apoptotic and anti apoptotic genes related to T cell apoptosis by fluorescence real time PCR in aging subjects.Results The percentage of T cell apoptosis from aging subjects after the treatment in the Kidney tonifying Group was significantly lower than that of the Placebo Group( P <0.01). Moreover, the expression of FasL and TNFR1 gene were downregulated significantly ( P <0.05), the expression of bcl 2 was upregulated significantly in the Kidney tonifying Group compared with that of the Placebo Group ( P <0.05). Conclusions Kidney tonifying recipe has the negative regulating effects on the transcription of pro apoptotic genes in aging subjects, at the same time, it also positively upregulates the transcription of anti apoptotic gene this kind of cooperating pattern is one of the molecular mechanisms underlying the effect of kidney tonifying recipe in downregulating the excessive T cell apoptosis in aging subjects.
