摘要
通过碱性培养基分离到耐碱的芽孢杆菌HSⅠ ,研究了碳源、氮源、培养温度、培养基起始 pH、培养时间、接种量、通气量、添加物对HSⅠ菌产木聚糖酶的影响 ,测定出其产酶最佳培养条件 :麸皮 6 %,(NH4 ) 2 SO4 0 .2 %,NaNO3 0 .4 %,蛋白胨 0 .6 %,起始 pH为 10 .5 ,接种量为 5 %,2 5 0mL三角瓶装液量为 5 0mL。最佳培养温度为 37℃ ,培养时间为 96h。其酶活力最高达 5 4IU/mL。
A strain producing xylanase was screened and identified as Bacillus sp.HSⅠ.The medium was composed of bran 6% ,(NH 4) 2SO 4 0.2%, NaNO 3 0.4% and peptone 0.6% .The optimum conditions for producing xylanase were initial pH 10.5,seed volume 5% and temperature 37℃.The fermentation time was 96h and the highest xylanase activity of 54IU/mL was obtained.
