摘要
采用 2 0 %胎牛血清 (FBS)、碱性成纤维细胞生长因子 (b FGF)所诱导的大鼠 VSMCs增殖模型 ,观察了海洋贝类提取物 (ESs)对血管平滑肌细胞 (VSMCs)增殖的影响。将培养的 5~ 8代细胞接种于 96孔板 ,除正常组加入含 0 .4 % FBS的 M199培养液外 ,其他组均加入含 2 0 % FBS培养液 (或含 5 0 ng/ ml b FGF的 0 .4 % FBS培养液 ) ,同时给药组分别加入含有 2 5、5 0、10 0 μl/ ml浓度的 ESs,孵育 4 8h后 ,以细胞计数、结晶紫染色及 MTT比色法测定 VSMCs生长情况。结果表明 ,与模型组比较 ,ESs各剂量组 VSMCs的增殖受到明显抑制 (P<0 .0 5、P<0 .0 1)。ESs对 2 0 % FBS和 b FGF所致的 VSMCs增殖具有明显抑制作用 。
The effects of the extract of seashells (ESs) on the proliferation of vascular smooth muscle cells (VSMCs) were investigated in vitro culture. 5~8 stage subcultured VSMCs were incubated into 96 well dish. VSMCs proliferation were produced with 20% FBS or 0 4% FBS containing 50 ng/ml bFGF, and by addition of 25?50?100 μl/ml ESs respectively,the inhibitory effects of ESs on the proliferation of VSMCs were determined by cell counting, crystal violet staining and MTT method after the incubation for 48 h. The results suggested that the proliferation of VSMCs in ESs groups were obviously inhibited, compared with model group ( P<0 05,P <0 01). ESs obviously inhibited the proliferation of VSMCs induced by 20% FBS and bFGF, which was associated with the concentrations.
