摘要
目的:应用人发角蛋白神经导管植人兔胫神经缺损部分,诱导神经再生过程中不同时期泛素(ubiquitin)的表达和分布。方法:免疫组织化学法。结果:术后48d,缝合线之间变性组织及其周围神经组织的雪旺细胞胞质和胞核中泛素免疫组化染色呈棕黄色阳性反应。术后99d,泛素在人发角蛋白周围的组织中免疫组化染色呈深棕色强阳性反应,在其它新生神经组织分布稀疏,免疫组化染色呈浅黄色弱阳性。术后99d较成熟的神经组织中无泛素阳性着色,而新生的神经组织呈棕黄色阳性反应。结论:人发角蛋白及缝合线之间变性组织的异常蛋白的降解主要由泛素承担。在新生神经组织中大量泛素参与蛋白质和变性神经组织的降解,而在趋于成熟的神经组织中因降解的蛋白质量的减少,参与降解的泛素量也减少。
Objective: To study the expression and distribution of ubiquitin during the repair period of damaged rabbit shank nerve induced by nerve conduit made of human hair keratin (HHK). Methods: Immuno - chemistry. Results: Ubiquitin in denatured tissue between sutures and cytoplasm, nuclei of schwann cells around them was dyed in brown - yellow, at 48 days after operation. Ubiquitin in tissue around HHK was dyed in nigger - brown, and that in other newly generated nerve tissue was brown and distributed sparsely. The positive rate of ubiquitin was very low in the nearly mature nerve tissue and high in newly generated nerve tissue, at 99 days after operation. Conclusions: Degeneration of HHK and abnormal proteins in denatured tissue between sutures may mainly be undertaken by ubiquitin. There is more ubiqitin that take part in clearance of abnormal proteins in newly generated nerve tissue, less in nearly mature nerve tissue.
