榄香烯乳对大鼠胃粘膜癌变c-myc基因表达及端粒酶活性的影响 被引量:8

Effects of Elemene on c-myc gene mRNA Expression and Telomerase Activity of Gastric Mucosa Carcinomatous Changes in Wistar Rats

摘要 目的:探讨榄香烯乳对大鼠胃粘膜癌变发生发展中c-myc基因表达及端粒酶活性的影响。方法:采用MNNG、10%NaCl混合液加酒精灌胃建立大鼠胃癌模型,设立正常组、模型组及榄香烯组,榄香烯组在造模过程中采用榄香烯乳干预。30周后,分别采用原位杂交、免疫组化及端粒酶微孔板杂交法检测大鼠胃粘膜c-myc基因mRNA、蛋白表达及端粒酶活性。结果:榄香烯组癌变发生率低于模型组。c-myc基因表达与端粒酶活性随着胃粘膜癌变的发展呈递增趋势,并且二者具有高度相关性。结论:下调c-myc基因表达,抑制端粒酶活性,是榄香烯乳重要的抗癌机制之一。 Objective:To study the effects of elemene on c-myc gene expression and telom erase activity of gastric cancer in Wistar rats.Methods:Thirty weeks after establishing the elemene groups with elemene in gastric cancer model induced by MNNG,10%sodium chlo ride and alcohol,we studied the expression of c-myc gene mRNA protein and telomerase activity us-ing in situ hybridization and immunohistochemistry and TRAP-microtiter plate hybridizationassay in rats in three groups:1.Normal control,2.Model control,3.Elemene group.Results:The rate of carcinogenesis of gastric cancer in elemene group was lower than those in the model groups.The expression of c -myc and telomerase activity escalated with the progression of can cerous changes in gastric mucosa and a close correlation was observed between them.Conclusion:El-emene can reduce the expression of c-myc gene and repress the telomerase activity of gastric cancer.
