摘要
目的探讨Wnt/β-catenin信号转导通路核心蛋白β-catenin在结肠癌组织中的异位表达情况及对结肠癌细胞增殖能力的影响。方法收集解放军武汉总医院病理科存档的结肠癌组织标本84例,免疫组织化学检测β-catenin蛋白的异位表达情况,并分析其异位表达与临床病理因素的关系。根据文献报道合成β-catenin siRNA,脂质体转染入结肠癌SW480细胞,验证其干扰有效后,细胞计数试剂盒8(cell counting kit 8,CCK-8)检测SW480细胞增殖。结果 84例结肠癌患者中,细胞浆或细胞核异位表达者64例,占76.19%。β-catenin蛋白的异位表达与肿瘤淋巴结转移状况和浸润深度相关。β-catenin siRNA抑制结肠癌SW480细胞增殖后,与空白对照组和阴性对照组比较,干扰组SW480细胞增殖能力显著下降。结论信号转导蛋白β-catenin在结肠癌细胞中存在细胞浆或细胞核的异位表达,并与肿瘤侵袭转移相关。特异性抑制β-catenin基因的表达可抑制结肠癌细胞的增殖能力。
Objective To explore the ectopic expression of β-catenin of the Wnt/β-catenin signal pathuay in colon cancer tissues and the effect of β-catenin on proliferation of colon cancer cells.Methods A total of 84 colon cancer patients who underwent surgery in the General Hospital of Central Theater Command were included.The ectopic expression of β-catenin was detected by immunohistochemistry,then the ectopic expression in patients of different clinicopathological factors were analyzed.The β-catenin siRNA was constructed and transfected into colon cancer SW480 cell by liposome,and the proliferation ability of SW480 cell was detected by cell counting kit 8(CCK-8)method after the effective interference.Results A total of 64 patients were β-catenin positive cases among 84 colon cancer patients,and the positive rate of ectopic expression was 76.19%.The ectopic expression of β-catenin on the cytoplasm and nucleus was found to be associated with lymph node metastasis and tumor invasion.The expression of SW480 cell was suppressed after β-catenin siRNAs intervention,SW480 cell proliferation of interference group was markedly inhibited than that of negative control group and blank group.Conclusion The ectopic expression of β-catenin on the cytoplasm or nucleus exists in colon cancer patients.The ectopic expression is found to be probably associated with metastasis and tumor invasion.Down regulation of β-catenin could decrease the proliferation ability of colon cancer cell.
