摘要
为研究鸡白痢沙门菌双组份系统CpxR/A的作用,本研究利用λ-Red重组技术构建了鸡白痢沙门菌cpxR基因缺失突变株ATCC19945ΔcpxR,通过应激试验和细胞感染试验,揭示cpxR基因在鸡白痢沙门菌致病机制中的重要作用。实验结果表明,cpxR基因缺失并未影响细菌的生长速度、生化特性和在鼠源巨噬细胞Raw264.7中的存活能力,但使得细菌对热、酸、碱和氧化应激的敏感性增加,同时在蛋清中的生存能力下降。本研究为进一步阐释CpxR/A系统的重要功能奠定基础。
To investigate the roles of two-component regulatory systems CpxR/A of Salmonella enterica serovar pullorum,the cpxR gene was knocked out from S.pullorum ATCC19945strain by λ-Red recombination,and examined by stress test and cell infection test for analysis its contribution to pathogenic mechanism of S.pullorum.The data demonstrated that the CpxR-deficient mutant showed no difference in growth characteristics,biochemical characteristics and survival ability in Raw 264.7,compared with wild type strain.However,the mutant strain showed increased sensibility to environment stress including heat,acid,alkali, oxidative and survival defection in albumen.This study provided the basis for exploring underlying mechanism of CpxR/A system.
