摘要
目的:探讨鹿角多肽对骨髓间充质干细胞(BMSCs)的影响。方法:利用HPLC法检测经超滤所得的不同分子量的鹿角多肽,并考察不同浓度、不同分子量的鹿角多肽对BMSCs增殖、生长周期、成骨化的影响。结果:BMSCs生长良好;当鹿角多肽(分子量800～1 500)浓度为1.261×10-1g/mL时,细胞增殖率45.84%,促进细胞增殖效果明显优于其余各浓度样品;鹿角多肽(分子量为800～1 500)可明显增加大鼠BMSCs的ALP值,与空白组比较具有显著性差异(P<0.01);流式细胞仪检测显示,鹿角多肽组(分子量800～1 500)对成骨细胞的增殖指数为38.68%,优于其他各组。结论:鹿角多肽(分子量800～1 500、浓度1.261×10-1g/mL)对BMSCs具有良好的促增殖、促成骨化作用。
Objective: To discuss the effects of antler polypeptide on bone marrow mesenchymal stem cells(BMSCs). Methods: HPLC method was used to detect different molecular weight antler polypeptide obtained by ultrafiltration. And the effects of different concentrations and different molecular weight antler polypeptide on the proliferation, growth cycle and osteogenesis of BMSCs were investigated. Results: BMSCs grown well. When the concentration of antler polypeptide(molecular weight 800～1 500) was 1.261×10-1 g/mL, the cell proliferation rate was 45.84%, and the cell proliferation was better than the other samples. Antler polypeptide(molecular weight 800～1 500) could significantly increased the ALP of rats bone marrow mesenchymal stem cells, which was significantly different from that of blank group(P<0.01). Flow cytometry showed that the antler polypeptide group(molecular weight 800～1 500) on osteoblast proliferation index was 38.68%, which was better than the other groups. Conclusion: Antler polypeptide(molecular weight 800-1 500, concentration of 1.261×10-1 g/mL) has a good effect on promoting proliferation and facilitating ossification of BMSCs.
