摘要
目的 :研究姜黄素 (Cur)诱导人胃癌BGC 82 3细胞凋亡的作用。方法 :细胞增殖抑制采用MTT法 ,细胞凋亡采用流式细胞仪检测。结果 :Cur 2 7~ 1 6 8μmol/L呈剂量依赖性抑制人胃癌BGC 82 3细胞增殖 (r =0 .994,P <0 .0 1 )。Cur 2 7μmol/L浓度开始出现BGC 82 3细胞Apo峰 ,6 8~ 1 36 μmol/L出现明显的Apo峰 ,细胞凋亡率在 2 4~ 72h内随作用时间延长逐渐增高。 结论 :姜黄素可诱导BGC 82 3细胞凋亡 ,该作用有时间和剂量依赖性。
AIM:To study the apoptotic induced by curcumin in huamn BGC-823 cells. METHODS:Inhibition of BGC-823 cells proliferation was measured using MTT assay. The amount of apototic cells was measured by flow cytometry. RESULTS:Curcumin 27~168μmol/L inhibited BGC 823 cell proliferation in a concentration-dependent manner(r=0.994,P<0.01)after 24h treatment with curcumin. Curcumin 27~168μmol/L elicited typical apoptosis of BGC 823 cell in a time-and concentration-dependent manner. About 15.94, 29.10 and 37.53% of the BGC-823 cells showed apoptosis after 24, 48 and 72h treatment with curcumin 27μmol/L respectively; and about 15.05, 29.68 and 53.09% cell apoptosis after 24, 48 and 72h treatment with curcumin 68μmol/L and about 17.47, 64.88, 84.09% cell apoptosis after 24, 48 and 72h treatment with curcumin 138μmol/L . CONCLUSION:[Curcumin induced apoptosis in BGC-823 cells in a time and concentration-dependent manner.
