摘要
目的 :探讨低血流心肌缺血促进葡萄糖摄取增加的机制。方法 :采用 Northern印迹法观察缺血心肌葡萄糖转运子 4( GLUT4) m RNA的表达 ,并利用蛋白质印迹法分析心肌 GL UT4多肽的表达。 结果 :局部心肌低血流缺血后 ,心肌 GL UT4m RNA和 GL UT4多肽表达明显增加 ;同时伴随缺血心肌葡萄糖摄取明显增多。 结论 :心肌缺血能刺激 GLU T4m RNA和GLU T4多肽表达 ,使 GLUT4数增加 ,进而促进心肌葡萄糖摄取增多 ,使缺血心肌能量需求得以平衡 ,有助于缺血心肌功能的恢复。提示低血流缺血刺激心肌 GL
Objective:To investigate the mechanism of low flow myocardial ischemia induced glucose uptake increase. Methods:The expression of GLUT4 mRNA was determined by semiquantitative northern blotting. The expression of myocardial GLUT4 polypeptide was determined by semiquantitative immunoblotting. Results:After 8 hour low flow myocardial ischemia, GLUT4 mRNA and GLUT4 polypeptide expression was significantly increased in ischemic regions in the experimental hearts. The glucose uptake was up regulated at the same time. Conclusion:Myocardial ischemia can stimulate the expression of GLUT4 mRNA and GLUT4 polypeptide in ischemic myocardial regions. Enhanced GLUT4 expression is very important for myocardial cells increasing glucose uptake during low flow myocardial ischemia.
