摘要
LAK细胞调节因素的研究对揭示LAK细胞的内在规律以及免疫过继疗法的开展与完善均具有重要意义。实验发现胰岛素能促进LAK细胞的增殖,提高细胞的产率,但是胰岛素不能替代IL-2使经短期IL-2刺激的淋巴细胞继续增殖。胰岛素还能促进LAK细胞表面IL-2R的表达。这可能是胰岛素促进LAK细胞增殖的重要机制。
As previously reported, the culture of mouse splenic cells cultured with the presence of human rIL-2 for 2-3 days would display generation of LAK cells which were capable of lysing a variety of tumor cells. Addition of insulin to the culture was found to enhance the proliferation of LAK cells without inhibition on the cytotoxicity.However, after lymphocytes were first activated by short term exposure to rIL-2, insulin could not be used to replace rIL-2 for maintaining proliferation. Studies using FACS (Becton-Dickson) analyzer revealed that insulin would enhance the surface expression of IL-2R on LAK cells which might be the possible mechanism of promoting proliferation.
