稀土元素对红豆杉细胞悬浮培养及紫杉醇合成的影响 被引量:5

摘要 研究了在 2 5 0mL摇瓶中 ,不同浓度的硝酸镧、硫酸铈铵、硝酸亚铈 3种稀土化合物对细胞生长及紫杉醇分泌和释放的影响。结果表明 ,在培养初期加入稀土元素 ,3种不同稀土化合物对细胞生长影响强弱不同 ,但趋势相似 ,均使细胞的延迟期缩短。 1ppm的Ce4 + 促进细胞生长的效果最明显 ,细胞干重第17d达到 10 9g/L。在指数期加入稀土元素 ,10ppmCe3 + 刺激细胞生长的效果最明显 ,细胞干重最高值达到 11 5g/L ,比对照高 1 5g/L ,而 10ppm的La3 + 抑制细胞的生长。经稀土元素处理后 ,细胞胞内和胞外紫杉醇含量都有大幅度的提高 ,其中以 10ppmCe3 + 处理 ,胞外紫杉醇释放率最大 ,达 37 7% The effects of three rare earth compounds including:Ce(NO\-3)\-2,Ce NH\-4(NO\-3)\-2,La NO\-3 on Taxus cell suspension culture and taxol synthesis were investigated in this paper.The results showed that the growth pattern of cell culture was altered by adding the rare earth elements in the medium at the beginning of culture.The lag phase was shortened and the exponential phase appeared earlier. The effection of lppm Ce 4+ on the improvement of cell growth was significant. The cell dry weight reached 10.9g/L. The growth of the cell was stimulated at lower concentration and inhibited at higher concentration if the rare earth elements were supplemented at the exponential phase. The cell dry weight could reach 11.5g/L when 10ppm Ce 3+ was added into the medium, which was higher 1.5g/l than control. The rare earth elements also affected the taxol synthesis and secretion. The results indicated that both the taxol contents in the cell and out the cell increased after add the rare earth elements in the medium. Supplemented 10ppm Ce 3+ at the beginning of cultre, the secretion rate of taxol reached maximun 37.7%.
