摘要
目的通过比较不同方法制备兔富血小板血浆,用CaCl2做诱导剂测量血小板计数、白细胞计数、血小板衍化生长因子(PDGF)及转化生长因子-β1(TGF-β1),探讨手工富血小板血浆制备方法及在CaCl2激活后的影响。方法采用一次离心法,二次离心法(Landesberg法和Aghaloo法)制备大耳白兔富血小板血浆进行比较。检测3组富血小板血浆中血小板计数、白细胞计数。用CaCl2做诱导剂,比较激活前后PDGF及TGF-β1的变化。结果 3种方法制备的富血小板血浆中血小板计数差异有统计学意义(P<0.01),Landesberg法和Aghaloo法均可制备有效浓度的富血小板血浆。一次离心法由于干扰较小,比较稳定,但血小板数不高;二次离心法富血小板血浆中血小板数相对高,手工制备容易受很多因素影响,且不稳定。3组富血小板血浆用CaCl2诱导前后,一次离心法、Aghaloo法PDGF与正常血浆组比较,差异无统计学意义(P>0.05)。Landesberg法制备的血小板血浆中PDGF、TGF-β1水平明显高于激活前(P<0.01)。一次离心法、Landesberg法和Aghaloo法富血小板血浆中血小板计数与白细胞计数呈正相关。结论手工制备兔富血小板血浆、Landesberg法和Aghaloo法都可以获得有效浓度,而Aghaloo法获得浓度最高。运用CaCl2做诱导剂,使富血小板血浆激活,可释放生长因子,起到治疗作用。
Objective In this assay, we measured the number of platelets, white blood cells, the amount of platelet derivation growth factor (PDGF), and transforming growth factor beta 1 (TGF-heta 1) with different methods in preparation of rabbit platelet-rich plasma, followed by CaC12 induction treatment to explore the differences among manual plateletrich plasma preparation methods and the influence of CaC12. Methods We used one-step centrifuge method and two-step centrifugation method (Landesberg and Aghaloo) to prepare big-ear rabbits platelet-rich plasma. We tested the number of platelets white blood cells in these 3 groups: The amount of growth factor (PDGF) and transforming growth factor beta 1 (TGF - beta i) changes before and after activation by CaCI2 were also tested. Results In this assay, three methods in preparation of platelet-rieh plasma showed significant difference (P〈0.01),Landesberg and Aghaloo method can effective obtain relatively high concentration of platelet rich plasma. One-step centrifuge method presented its low interferes and more stable advantage. However, the ptatelet counts were not high. Using Two-step eentrifugation the prepared platelet-rieh plasma was relatively high, however manual preparation was influenced by many factors, which was not stable. Three groups were compared before and after platelet rich plasma induction by CaC12 .. there was no significant difference in platelet derivation growth factor between one-step centrifuge method, Aghaloo method and normal plasma (P〉 0. 05 ). Using Landesberg method the platelet plasma derivation growth factors, transforming growth factor beta 1 (TGF-beta 1) level were significantly higher than the state of activation (P〈0.01). The platelets and white blood cell counts in one-step centrifuge method, Landesberg method and Aghaloo method were positively correlated. Conclusion In Manual preparation of rabbit platelet-rich plasma, Landesberg and Aghaloo method can get effective concentration, while Aghaloo method can obtain the highest concentration. Activating the piatelet-rich plasma by CaC12 can release the growth factors and it is effective.
